摘要 目的:评价经腺病毒介导的血管内皮细胞生长因子(VEGF)基因转染后的大鼠骨髓基质干细胞(BMSCs)诱导成内皮细胞的可行性。方法:从大鼠骨髓中分离培养获取BMSCs,分别经Adv-VEGF、Adv-GFP转染,Western印迹分析检测VEGF蛋白的表达情况,细胞生长曲线测定BMSCs增殖特性,同时进行免疫表型鉴定,并与未转染细胞组比较。结果:AdvVEGF转染组BMSCs可分泌VEGF蛋白,而其余两组不分泌VEGF蛋白。转染VEGF基因后,BMSCs生长速度变快,向内皮细胞分化。与Adv-VEGF转染组BMSCs相比,Adv-GFP转染细胞组、未转染细胞组的BMSCs生长速度较慢,未有向内皮细胞分化的证掘。结论:腺病毒介导的VEGF基因转染可诱导BMSCs分化成内皮细胞。 Objective: To investigate the feasibility of using vascular endothelial cell growth factor (VEGF) gene to in duce hone marrow stromal stem cells(BMSCs) into endothelial cells in vitro. Methods :BMSCs were isolated from mouse bone marrow and cultured in vitro. They were divided into 3 groups : Adv- VEGF transfected group, Adv-GFP transfected group and un-transfeeted group. Western blot analysis .cell growth curves determination, and immunohistochemistry were performed. Results: After induced with Adv-VEGF, BMSCs grew faster and differentiated into endothelial cells. Compared with the Adv-VKGF transfected group. BMSCs in other groups grew slowly and didnot differentiate into endothelial cells. In Adv-VKGF transfected group. Western blot analysis showed that these cells indeed produced the VEGF protein,but Adv-GFP transfected group and un transfected group didnot. Conclusion : Adenovirus mediated VEGF gene transfection can induce endothelial cell from BMSCs.
