摘要 目的 研究胰腺癌组织及细胞中Bax/Bcl-2蛋白和FHIT mRNA的表达,探讨FHIT与凋亡相关因子在胰腺癌中的关系。方法 应用ABC免疫组织化学方法检测胰腺癌细胞和组织标本中Bax与Be1-2蛋白,RT-PCR检测其中FHIT mRNA的表达。结果 38例胰腺癌中Bax蛋白阳性17例(45%),Bcl-2蛋白阳性表达22例(58%),FHIT mRNA异常表达为21例(56%)。转染有外源性FHIT的1990细胞(1990 pFHIT)Bax表达阳性率有升高,Bcl-2/Bax比值下降。结论 Bax、Bcl-2及FHIT都通过调控凋亡参与胰腺癌的病理过程。FHIT诱导凋亡可能与Bax上调表达有关。 Objective To study the expression of Bax, Bcl-2 protein and FHIT mRNA in pancreatic carcinoma and explore the significance of their expression in the process of pancreatic carcinoma. Methods Bax, Bcl-2 protein was detected by immunohistochemistry of avidin-biotin complex, and FHIT mRNA was checked by RT-PCR. Results The positive rate of Bax and Bcl-2 protein was 45% and 58%, respectively, in 38 cases of pancreatic carcinoma, and the negative expressing rate of FHIT mRNA was 56%. Immunohistochemical study revealed that the expression of Bax was increased after 1990 cell line transfected with FHIT gene (named as 1990 pFHIT cells). Conclusions Bax,Bcl-2 and FHIT are involved in the development of pancreatic carcinoma by adjusting the path of apoptosis, and the apoptosis induced by FHIT may be related to Bax protein.
