摘要
本研究旨在探讨止颤控涎丸(ZCKXW)对帕金森病(Parkinson's disease,PD)模型小鼠的神经保护作用。试验建立C57BL/6N小鼠PD模型,随机分为PD模型组、盐酸金刚烷胺组及ZCKXW高、低剂量组,分别灌胃给予纯净水、19.29 mg/kg、2.73 g/kg、1.365 g/kg,空白组灌胃等体积纯净水,连续灌胃15 d,末次给药后进行行为学试验并收集标本,检测小鼠黑质中酪氨酸羟化酶(Tyrosine Hydroxylase,TH)、前列腺素E2(Prostaglandin E2,PGE2)含量,通过Western blot检测纹状体中α-突触核蛋白(α-synuclein,α-Syn)和环氧化酶-2(Cyclooxygenase-2,COX-2)蛋白表达水平。与模型组比较,ZCKXW各剂量组均可显著缩短PD模型小鼠的爬杆时间(P<0.05,P<0.01),增加疲劳转棒时间(P<0.05);ZCKXW各剂量组均能降低黑质中PGE2含量,但无显著性差异;ZCKXW各剂量组均能显著提高TH水平(P<0.05);ZCKXW高剂量组可显著下调小鼠纹状体中α-Syn和COX-2蛋白表达(P<0.01),ZCKXW低剂量组可明显降低纹状体中COX-2蛋白的表达水平(P<0.05)。ZCKXW通过上调TH表达,降低炎症因子的生成,减少了多巴胺能神经元的损伤,改善了PD的症状,为治疗帕金森症提供了一种新的思路和方法。
The aim of this study is to investigate the neuroprotective effect of Zhichankongxianwan(ZCKXW)on the mouse model of Parkinson's disease(PD).To establish PD model in C57BL/6N mice.After the modeling was successful,they were randomly divided into the model group,the amantadine hydrochloride group,and the high and low dose groups of ZCKXW,and were given pure water,19.29 mg/kg,2.73 g/kg,and 1.365 g/kg.In the blank group,the same volume of purified water was given intragastrically.The mice were continuously given intragastrically for 15 days.After the last administration,behavioral experiments were performed and specimens were collected,and the content levels of tyrosine hydroxylase(TH)and prostaglandin E2(PGE2)in the mouse substantia nigra were detected,and the protein expression of alpha-synuclein(α-Syn)and cyclooxygenase-2(COX-2)in the striatum was detected by Western blot.Compared with the model group,ZCKXW could significantly shorten the time of climbing the rod(P<0.05,P<0.01)and increase the time of fatigue rotation(P<0.05);ZCKXW could decrease the content of PGE2 in substantia nigra and significantly increase TH level in all dose groups(P<0.05),but there was no significant difference;the expression ofα-Syn and COX-2 protein in the striatum of mice was significantly down-regulated in the high-dose group of ZCKXW(P<0.01),and the expression of COX-2 protein in the striatum was significantly down-regulated in the low-dose group of ZCKXW(P<0.05).ZCKXW reduces the damage of dopaminergic neurons and improves the symptoms of PD by up-regulating the expression of TH and reducing the production of inflammatory factors.It provides a new idea and method for the treatment of Parkinson's disease.
