摘要
多糖是中药煎剂的重要组成成分,具有调节免疫、抗氧化、抗炎等广泛的药理活性。多糖的活性与分子量、单糖组成、糖残基的连接方式、取代基结构类型等信息息息相关。多糖作为复杂的高分子化合物,结构解析困难,且难以形成结晶,作用靶点不清,构效关系难以阐明。因此系统表征多糖的化学结构,以便更好地开发利用中药多糖是亟待开展的重要工作。现代分析技术如色谱技术、质谱技术、光谱技术以及核磁共振波谱技术等已成为解析中药多糖结构的重要手段,本文系统综述了这些仪器分析技术在多糖分子量、单糖组成、糖苷键的连接方式以及高级结构测定等方面的应用,以期为多糖结构的解析提供参考。
Polysaccharides are important constituents of herbal decoctions and have a wide range of pharmacological activities such as immune regulation,antioxidation and anti-inflammation.The activity of polysaccharides is closely related to the molecular weight,monosaccharide composition,linkage of sugar residues,substituent structure type and other information.As complex macromolecular compounds,polysaccharides are difficult to characterize structurally,in addition,polysaccharides are difficult to form crystals,the target of action and structure-activity relationship are difficult to elucidate.Therefore,it is an urgent task to systematically characterize the chemical structures of polysaccharides to better exploit polysaccharides.Modern analytical techniques such as chromatography,mass spectrometry,specturm,and nuclear magnetic resonance spectroscopy have become important tools in analyzing the structure of polysaccharides.This paper systematically reviewed these instrumental analytical techniques in the determination of molecular weight,monosaccharides composition,linkage of glycosidic bonds and senior structure,aiming to provide reference for the analysis of polysaccharide structure.
