摘要
目的:安徽霍山为兰科石斛属植物地理分布的最北端,主产3种石斛:霍山石斛、细茎石斛和铁皮石斛,其多糖提取物具有显著的药理活性,本研究旨在探究同一特殊生境下3种石斛所含多糖在组成上的相似和差异。方法:实验以越冬茎条为供试材料,采用苯酚-硫酸法分析多糖含量差异,利用HPSEC法分析分子量分布差异,通过柱前衍生化HPLC法分析单糖组成差异。结果:霍山石斛、细茎石斛和铁皮石斛多糖含量分别为其干重的(18.65±1.50)%、(11.32±1.30)%和(20.78±1.60)%;其多糖分子量呈连续分布状态,其中分子量介于50~200 kDa的高活性多糖在3种石斛多糖中的比例分别为:(16.38±1.40)%、(21.61±1.30)%和(14.19±1.50)%;3种石斛多糖水解产物中检出葡萄糖(Glc)、甘露糖(Man)、半乳糖(Gal)、木糖(Xyl)、葡萄糖醛酸(GlcUA)、半乳糖醛酸(GalUA)、阿拉伯糖(Ara)共7种单糖,且都以甘露糖和葡萄糖为主要单糖组分,但铁皮石斛中的甘露糖和葡萄糖的相对峰面积比值最大。结论:同一生境下3种石斛所含多糖在含量、分子量分布和单糖组成上既存在共性也存在明显差异,实验探究了同一特殊生境下3种石斛多糖组分间的异同,其结果对石斛的鉴别和组效关系的研究提供了重要参考。
Objective:Huoshan in Anhui was the extreme north of geographic distribution for Dendrobium plants,produced three kinds Dendrobium plants,Dendrobium huoshanense,Dendrobium moniliforme and Dendrobium officinale,their polysaccharide extract had significant pharmacological activity,this study aimed to explore the similarities and differences on the polysaccharide composition of three kinds of Dendrobium plants in the same habitat.Methods:Wintering stem was selected to tested material,differences of content of three kinds of Dendrobium polysaccharides were analyzed by phenol-sulfuric acid method,differences of molecular weight distribution of three kinds of Dendrobium polysaccharides were analyzed by high performance size exclusion chromatography(HPSEC).Differences of monosaccharide composition of three kinds of Dendrobium polysaccharides were analyzed by column derivatization HPLC.Results:The polysaccharides content accounted for(18.65 ± 1.50)%,(11.32 + 1.30)% and(20.78 + 1.60) % of dry weight,respectively in Dendrobium huoshanense,Dendrobium moniliforme and Dendrobium officinale,and their molecular weight distribution range overlap each other,and the proportion of highly active polysaccharide whose molecular weight between 50 ~200 kDa in three kinds of Dendrobium polysaccharides was(16.38 + 1.40) %,(21.61 ± 1.30) % and(14.19 ± 1.50) %,respectively.Total seven monosaccharides including glucose (Glc),mannose (Man),galactose (Gal),xylose (Xyl),glucuronic acid (GlcUA),galacturonic acid (GalUA) and Arabian sugar (Ara),were detected in polysaccharide hydrolysate of three kinds Dendrobium,and mannose and glucose were all the main monosaccharide components,but the relative peak area ratio of mannose and glucose of Dendrobium officinale was the largest.Conclusion:The content and molecular weight distribution and monosaccharide composition of three kinds of Dendrobium polysaccharide have both similarities and obviously differences in the same habitat.The experiment results provide important reference for identifying and exploring the quality differences,especially the group activity relationship,of three kinds of Dendrobium species.
