益气活血开心汤改善冠心病合并抑郁大鼠抑郁样行为作用机制研究

Study on the Mechanism of Yiqi Huoxue Kaixin Decoction(益气活血开心汤)in Improving Depression like Behavior in Rats with Coronary Heart Disease and Depression

摘要 目的探讨益气活血开心汤是否通过p-CREB/BDNF途径改善大鼠冠心病(CHD)后抑郁症状以及是否能够抑制海马神经元的凋亡。方法将SD大鼠随机分为4组:空白组、模型组、中药组、西药组。采用结扎冠状动脉左前降支构建急性心肌梗死(AMI)模型,联合慢性不可预知刺激(CUMS)21 d构建CHD合并抑郁模型;同时,中药组、西药组分别给予益气活血开心汤、氟西汀灌胃21 d。通过大鼠整体状态、体质量、旷场实验、强迫游泳和蔗糖偏好实验评估抑郁样行为;尼氏染色观察海马神经元的形态结构和凋亡情况;Western blot和免疫荧光评估海马复合体CA1区磷酸化的环磷酸腺苷反应元件结合蛋白(p-CREB)和脑源性神经营养因子(BDNF)的表达。结果大鼠的抑郁样行为经过益气活血开心汤和氟西汀治疗后得到明显改善(P<0.05,P<0.01);海马CA1区的p-CREB和BDNF的表达明显增多(P<0.05)。结论结果表明,益气活血开心汤可以明显改善CHD后抑郁大鼠的抑郁症状和神经功能,并能够抑制海马神经元的凋亡情况,且与氟西汀的改善作用无显著差异。 Objective To explore whether Yiqi Huoxue Kaixin Decoction(益气活血开心汤)can improve depression symptoms after coronary heart disease(CHD)in rats through the p-CREB/BDNF pathway and whether it can inhibit apoptosis of hippocampal neurons.Methods SD rats were randomly divided into four groups:blank group,model group,traditional Chinese medicine group,and Western medicine group.Using ligation of the left anterior descending coronary artery to construct an acute myocardial infarction(AMI)model combined with chronic unpredictable stimulation(CUMS)for 21 days to construct a CHD combined with depression model;Meanwhile,the traditional Chinese medicine group and the Western medicine group were respectively given Yiqi Huoxue Kaixin Decoction and fluoxetine by gavage for 21 days.To evaluate the overall state,weight,open field experiments,forced swimming,and sucrose preference experiments of rats to assess depressive like behavior;Nissl staining was used to observe the morphological structure and apoptosis of hippocampal neurons;Western blot and immunofluorescence were used to evaluate the expression of p-CREB and brain-derived neurotrophic factor(BDNF)in the CA1 region of the hippocampal complex.Results The depression like behavior of rats were significantly improved after treatment with Yiqi Huoxue Kaixin Decoction and fluoxetine(P<0.05,P<0.01);The expression of p-CREB and BDNF in the hippocampal CA1 region were significantly increased(P<0.05).Conclusion The results showed that Yiqi Huoxue Kaixin Decoction could significantly improve the depressive symptoms and neurological function of post CHD depression rats,and inhibit the apoptosis of hippocampal neurons,with no significant difference in improvement compared to fluoxetine.
