痰热清注射液和雾化吸入液治疗慢性阻塞性肺疾病模型大鼠药效比较研究 被引量:1

Comparative Study on the Efficacy of Tanreqing Injection(痰热清注射液)and Atomized Inhalation in the Treatment of Chronic Obstructive Pulmonary Disease Model Rats

摘要 目的比较痰热清注射液和雾化吸入液治疗慢性阻塞性肺疾病(简称慢阻肺)模型大鼠的作用效果。方法Wistar大鼠分为对照组、模型组、阳性组、注射液组和雾化组(高、中、低剂量组),采用细菌脂多糖(LPS)联合烟熏建立慢阻肺动物模型,造模和给药1个月后测定动物肺功能,检测肺组织的细胞因子,对肺组织进行HE组织病理学染色、Masson染色和糖原染色(PAS),分析各组肺泡的平均线性截距(MLI)、气道壁厚度和PAS阳性细胞数。结果慢阻肺模型组动物出现不同程度的咳嗽、喷嚏,严重者会有呼吸急促、鼻分泌物增加的现象,阳性组,注射液组和雾化吸入中、高剂量组的动物状态较模型组有明显的好转。肺功能结果显示雾化吸入高剂量组和中剂量组的动态顺应性(Cydn)和气道阻力(RI)较注射液组有明显的好转。HE结果显示,阳性组,注射液组,雾化吸入高剂量组和中剂量组动物肺组织肺泡结构相对完整,血管扩张有所改善。与注射液组比较,雾化吸入高剂量组和中剂量组的细胞因子表达、MLI、气道壁厚度、PAS阳性细胞数均有所降低,且差异具有统计学意义。结论痰热清雾化吸入制剂在治疗慢阻肺方面较注射液具有剂量小、效果明显的特色和优势。 Objective To compare the effects of Tanreqing Injection(痰热清注射液)and atomized inhalation in the treatment of chronic obstructive pulmonary disease(COPD)model rats.Methods Wistar rats were divided into control group,model group,positive group,injection group and atomization group(high,medium and low).The animal model of COPD was established by LPS combined with smoking.After modeling and administration for one month,the animal lung function was measured,and the cytokines in lung tissue were detected.The lung tissue was stained by HE histopathology,Masson staining and periodic acid schiff stain(PAS),and the average linear intercept(MLI)of alveoli in each group was analyzed.Results In the COPD model group,the animals had different degrees of cough and sneezing,and in severe cases,they had shortness of breath and increased nasal secretions.The animals in the positive group,the injection group and the middle and high doses of atomized inhalation group were obviously better than those in the model group.The results of lung function showed that the dynamic compliance(Cydn)and airway resistance(RI)in high and middle dose groups were significantly better than those in injection group.HE results showed that the alveolar structure of lung tissue in positive group,injection group,high dose group and middle dose group was relatively complete,and the vasodilation was improved.Compared with the injection group,the expression of cytokines,MLI,airway wall thickness and PAS positive cells in the high-dose group and the middle-dose group decreased,and the difference was statistically significant.Conclusion Tanreqing Aerosol Inhalation Preparation had the characteristics and advantages of small dosage and obvious effect compared with injection in the treatment of COPD.
