一种功能性奶粉对生长期大鼠骨健康的影响

Effects of a functional milk powder on bone health in growing rats

摘要 为评估一种添加了骨碎补的功能性奶粉对生长期大鼠骨健康的影响。将50只断乳SPF级雌性SD大鼠随机分为低钙对照组、碳酸钙对照组和奶粉低中高3个剂量组,灌胃90 d,于干预期结束前3 d进行钙代谢实验。干预结束后取双侧股骨进行股骨常规指标、骨密度、骨钙测定及抗酒石酸碱性磷酸酶免疫组化染色,并进行Micro-CT扫描骨微观结构,同时取血清测定I型胶原C端交联肽(C-telopeptide of typeⅠcollagen,CTX-Ⅰ)、Ⅰ型前胶原氨基端原肽(procollagen typeⅠamino-terminal peptide,PINP)和骨钙素(osteocalcin,BGP)的水平。结果表明,与低钙对照组相比,碳酸钙对照组和奶粉各剂量组的股骨干重和股骨长均显著升高(P<0.05,P<0.01),骨密度和骨钙含量均显著升高(P<0.01),CTX-Ⅰ、PINP及BGP水平均显著升高(P<0.01),其中高剂量组PINP/CTX-I比例显著降低(P<0.05);除奶粉低剂量组外,破骨细胞数量均显著降低(P<0.05,P<0.01);奶粉低和中剂量组的钙吸收率和钙储留率显著升高(P<0.01)。Micro-CT提示各奶粉剂量组的骨微观结构均优于低钙对照组。因此,受试奶粉具有有助于改善骨密度的作用,其机制可能与促进骨形成有关。 To evaluate the effects of a functional milk powder added with Drynariae Rhizoma on bone health in growing rats.50 weaned SPF-grade female SD rats were randomly divided into five groups:low-calcium control group,positive control group,low-dose milk powder group,medium-dose milk powder group,and high-dose milk powder group,gavaged for 90 days.A 3-day calcium-balanced experiment was performed before execution.Bilateral femurs were taken for measuring femur dry weights and lengths,bone mineral density,bone calcium levels,tartrate-resistant acid phosphatase staining,and micro-CT scanning.Besides,the serum levels of C-telopeptide of typeⅠcollagen(CTX-Ⅰ),PINP,and osteocalcin(BGP)were detected.Results have shown that compared with the low-calcium control group,the femur dry weight and femur length(P<0.05,P<0.01),the bone mineral density and bone calcium content(all P<0.01),and the serum levels of CTX-Ⅰ,procollagen typeⅠamino-terminal peptide(PINP),and BGP(all P<0.01)significantly increased in the positive control group and all milk powder dose groups;the number of osteoclasts was significantly reduced in all groups except for the low-dose milk power group,among which the PINP/CTX-I ratio in the high-dose milk power group was significantly decreased(P<0.05);except for the low-dose milk powder group,the number of osteoclasts was significantly decreased(P<0.05,P<0.01);the calcium absorption rate and calcium retention rate in the milk powder low and medium dose groups significantly increased(all P<0.01).Micro-CT showed that the bone microstructure of each milk powder dose group was better than that of the low-calcium control group.Thus,the milk powder could improve the bone health of growing rats by prompting bone formation.
