某市29批市售党参药材饮片高效液相色谱指纹图谱建立及化学计量学研究 被引量:4

Establishment of HPLC Fingerprint and Chemometrics Study of 29 Batches of Commercial Codonopsis Radix Decoction Pieces in a City

摘要 目的为市售党参药材饮片质量评价方法的建立提供参考。方法选取太原市市售29批党参药材饮片(来自11个厂家)作为样品,采用高效液相色谱(HPLC)法建立其HPLC指纹图谱,色谱柱为WatersXSelect®HSST3柱(250 mm×4.6 mm,5μm),流动相为0.5%磷酸水溶液-乙腈(梯度洗脱),流速为1.0 mL/min,检测波长为220 nm,柱温为35℃,进样量为10μL。使用中药色谱指纹图谱相似度评价系统(2012版)进行相似度评价,并进行聚类分析和主成分分析。结果29批样品的HPLC指纹图谱中共确定18个共有峰,并指认出其中3个共有峰,分别为党参苷Ⅰ峰(7号峰)、党参炔苷宁峰(10号峰)和党参炔苷峰(11号峰);29批样品中有28批相似度大于0.85,可聚为3类(分类距离为5);特征值排名前5成分的累积方差贡献率>80%,为样品主成分;载荷图显示,1-4、9-11,5-8、12,1、13-14,15-16,17-18峰分别对主成分1,2,3,4,5的贡献较大;综合得分介于-90.17~80.15分。结论太原市市售不同厂家的29批党参药材饮片质量差异较大。HPLC指纹图谱可较全面地表征党参药材饮片的化学质量特征,结合化学计量学方法,可为其质量评价提供参考。 Objective To provide a reference for the establishment of a quality evaluation method of commercial Codonopsis Radix Decoction Pieces.Methods A total of 29 batches of commercial Codonopsis Radix Decoction Pieces from 11 manufacturers in Taiyuan were selected as samples,and high-performance liquid chromatography(HPLC)method was used to establish the HPLC fingerprint.The chromatographic column was Waters XSelect®HSS T3 column(250 mm×4.6 mm,5µm),the mobile phase was 0.5%phosphoric acid aqueous solution-acetonitrile(gradient elution),the flow rate was 1.0 mL/min,the detection wavelength was 220 nm,the column temperature was 35℃,and the injection volume was 10µL.The Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine(Version 2012)was used for similarity evaluation,the cluster analysis and principal component analysis(PCA)were performed.Results A total of 18 common peaks were marked in the HPLC fingerprint of 29 batches of samples,and three common peaks were identified,including tangshenosideⅠpeak(peak 7),lobetyolinin peak(peak 10)and lobetyolin peak(peak 11).The similarity of HPLC fingerprint of 28 batches of samples was greater than 0.85,and 29 batches of samples could be clustered into three groups(with a classification distance of 5).The top five components with a high eigenvalue had a cumulative variance contribution rate greater than 80%,which were the principal components of the samples.The loading plot showed that the peaks 1-4,9-11(principal component 1),peaks 5-8,12(principal component 2),peaks 1,13-14(principal component 3),peaks 15-16(principal component 4)and peaks 17-18(principal component 5)contributed significantly to different principal components respectively.The comprehensive score of the principal components of 29 batches of samples was in the range of-90.17 to 80.15 points.Conclusion The quality of 29 batches of commercial Codonopsis Radix Decoction Pieces from different manufacturers in Taiyuan varies greatly.The HPLC fingerprint can comprehensively reveal the chemical quality characteristics of Codonopsis Radix Decoction Pieces,which can provide a reference for the quality evaluation of this medicinal herb combined with chemometrics analysis.
