两种听力老化小鼠听力特征及优缺点分析 被引量:2

摘要 目的利用听性脑干反应(ABR)阈值作为检测项目,系统总结C57BL/6J(B6J)小鼠与SAMP8小鼠在不同月龄时听力动态变化规律及其作为老年性听力损失研究模型的特点,并对比不同月龄C57BL/6J小鼠与SAMP8小鼠ABR反应阈值差异,为老年性听力损失研究动物模型选择提供经验。方法选用67只不同月龄C57BL/6J小鼠以及15只SAMP8小鼠,采用ABR阈值测试方法,以引出可辨认的ABR波I波的最小声强定为阈值,统计ABR反应阈值结果。结果C57BL/6J小鼠与SAMP8小鼠均表现出听力早衰问题。其中C57BL/6J小鼠在6月龄便有听力下降表现,但以高频听力衰退为主,直至18月龄时,出现双侧对称性全聋。该品系小鼠在听力衰老过程中表现出个体差异较大的特征,部分小鼠表现出双侧听力不对称性损失。SAMP8小鼠在5月龄时仅表现出高频听力损失,在7月龄时表现出全频听力下降,以高频听力损失为著。该品系小鼠个体差异较小,且表现为双侧听力对称性损失。结论C57BL/6J小鼠和SAMP8小鼠均可作为老年性听力损失研究的动物模型。C57BL/6J小鼠的优点在于:获取方式简单,对麻醉耐受性较好。缺点在于:该品系小鼠个体差异较大,部分小鼠在听力衰退过程中表现出双侧非对称性听力下降。SAMP8小鼠的优点在于:该品系小鼠个体差异较小,在听力衰退过程中表现为对称性听力下降。缺点在于:该品系小鼠麻醉耐受度较差,使用1%戊巴比妥麻醉易死亡,且引起听力早衰的作用机制不明确。 Objective To summarize and compare auditory brainstem response(ABR)thresholds in C57BL/6J and SAMP8 mice models of dynamic hearing changes at different months and their differences in studying senile to discuss their advantages and disadvantages.Method ABR thresholds were recorded in 67 C57BL/6J and 15 SAMP8 mice of different ages.Results Both C57BL/6J mice and SAMP8 mice showed early onset presbycusis,mainly involving high frequency at the age of 6 months and leading to bilateral symmetric total deafness at 18 months for C57BL/6J mice.Excluding the possibility of otitis media,C57BL/6J mice showed significant individual differences in the development of presbycusis,with some showing asymmetric hearing loss.SAMP8 mice showed only high-frequency hearing loss at 5months,followed by full-frequency loss at 7 months,although still with high-frequency loss as the primary feature.In addition,when otitis media was excluded,SAMP8 mice showed little individual differences and usually symmetrical hearing loss.Conclusion Both C57BL/6J mice and SAMP8 mice can be used as animal models of presbycusis.The advantage of C57BL/6J mice is that they are the most commonly used animal model for presbycusis research and the gene mutation site causing premature presbycusis is better known.They are easy to acquire,tolerate anesthesia well and usually survive hearing tests.The disadvantage of this strain include signficant individual differences and asymmetric hearing loss in some mice.In contrast,minimal individual variation is an advantage of the SAMP8 mice together with their usually symmetrical hearing.Their disadvantages involve poor anesthesia tolerance(1%pentobarbital can result in death)and unclear mechanisms of their hearing failure.
