淫羊藿苷对SAMP8小鼠肝脏、肾脏及胸腺的保护作用 被引量:5

摘要 目的观察淫羊藿苷(Icariin,ICA)对快速衰老小鼠(SAMP8)的肝脏、肾脏及胸腺的保护作用,并探索其可能机制。方法随机选取SAMR1小鼠作为对照组,雄性SAMP8小鼠随机分为4组(每组12只),SAMP8模型组及ICA(20、40、80 mg/kg)灌胃给药组,每天1次,连续3个月;SAMP8组和SAMR1组小鼠灌胃等体积的双蒸水,直至8月龄处死,取肝脏、肾脏及胸腺。β-半乳糖苷酶染色法对肝脏和肾脏进行染色;生化法检测肝脏、肾脏及胸腺匀浆上清液中丙二醛(MDA)的含量、超氧化物歧化酶(SOD)及谷胱甘肽过氧化物酶(GSH-Px)的活性。结果与SAMR1组相比,肝脏和肾脏的β-半乳糖苷酶染色发现SAMP8的蓝色颗粒较多;肝脏、肾脏及胸腺匀浆上清液中,SAMP8的MDA含量增多,而SOD和GSH-Px的活性减弱;与SAMP8组相比,ICA给药组β-半乳糖苷酶染色蓝色颗粒减少;MDA的含量减少,而SOD和GSH-Px的活性增加。结论 ICA具有保护肝脏、肾脏及胸腺的作用,其可能的机制与体内β-半乳糖苷酶、SOD、GSH-Px及脂质过氧化物有关。 Objective To investigate the protection of Icariin( ICA) on of liver,kidney and thymus in senescence accelerated mouse prone 8( SAMP8) mice and further explore the possible underlying mechanisms.Methods SAMP8 mice were randomly divided into model group,ICA low-,middle- and high- dose of ICA groups. whereas SAMR1 mice selected as control group. Low-,middle- and high- dose of ICA group were administered with ICA at doses of 20,40 and 80 mg / kg for consecutive 3 months. Control and model groups were administered with double- distilled water. Then,mice were sacrificed and liver,kidney and thymus were collected. β- galactosidase staining were performed to observe the number of aging cells in the liver and kidney.Moreover,the biochemical method was used to detect the content of malondialdehyde( MDA) and the activities of glutathione peroxidase( GSH- Px) and superoxide dismutase( SOD) in the liver,kidney and thymus homo genates. Results Compared with the control group,the mean number of blue particles and the content of MDA were significantly increased in the liver and kidney and the activities of GSH- Px and SOD were dramaticlly decreased in liver,kidney and thymus homogenates of model group. However,the mean number of blue particles and the content of MDA significantly were decreased,and the activities of GSH- Px and SOD were markedly increased in liver,kidney and thymus homogenates of ICA treatment group. Conclusion ICA could protect liver,kidney and thymus in SAMP8 mice,and the possible mechanisms might be related to the activity of SA- β-gal,SOD,GSH- Px and lipid peroxide.
