基于GC-MS和网络药理学研究南柴胡挥发油抗抑郁作用机制 被引量:5

Study on the Antidepressant Mechanism of the Essential Oil in Bupleurum scorzonerifolium Based on the GC-MS and Network Pharmacology

摘要 本研究基于气相色谱制。运用GC-MS技术,结合文献,对南柴胡挥发油化学成分进行分析鉴定;查找SwissADME数据库筛选满足“类药五原则”的成分;通过Swiss TargetPrediction数据库预测成分靶点,利用GeneCards和DrugBank数据库富集抑郁相关靶点;进一步使用Venny 2.1.0在线工具获取成分和疾病的交集靶点;在Cytoscape软件中构建PPI蛋白相互作用网络,筛选核心靶点;将核心靶点导入DAVID数据库完成GO和KEGG分析,随后使用微生信平台进行可视化分析并利用Cytoscape软件构建成分靶点通路图。结果从南柴胡挥发油中共鉴定出28个主要成分,且均满足“类药五原则”,最终筛选出ESR1、GSK3B、JUN、CXCL8、MMP2、F2R、GNAI3和PRKCA等45个核心靶点,富集分析发现南柴胡挥发油抗抑郁作用主要涉及的通路包括雌激素信号通路、Rap1信号通路、Reaxin信号通路、PI3K-Akt信号通路和cAMP信号通路等。本研究证明南柴胡挥发油抗抑郁作用可能是通过多靶点、多成分起作用,可为南柴胡药材质量标准的制订和提升提供更加可靠的依据。 This study aims to explore the material basis and potential mechanism of the antidepressant effect of the volatile oil in Bupleurum scorzonerifolium based on the gas chromatography-mass spectrometry(GC-MS)and network pharmacology.The chemical constituents of the essential oil from B.scorzonerifolium were analyzed by GC-MS combined with the literature.The components meeting the"five principles of drug class"were screened by searching the SwissADME database and the potential component targets were predicted through the SwissTargetPrediction database.GeneCards and DrugBank databases were used to enrich depression-related targets.The Venny 2.1.0 online tool was applied to obtain intersection targets of components and diseases.PPI protein interaction network was built through Cytoscape software to screen core targets.Import the core targets into the DAVID database to complete GO and KEGG analysis,and then use the microbiota platform for visual analysis and build a component target path diagram in Cytoscape software.As a result,a total of 28 main components were identified from the essential oil of B.scorzonerifolium,and all of them met the"five drug-like principles".Moreover,45 core targets,including ESR1,GSK3B,JUN,CXCL8,MMP2,F2R,GNAI3,and PRKCA,were screened.Enrichment analysis showed that the main pathways involved in the antidepressant effect of B.scorzonerifolium volatile oil included estrogen signal pathway,Rap1 signal pathway,Reaxin signal pathway,PI3K-Akt signal pathway,and cA MP signal pathway.This study proved that the anti-depression effect of the volatile oil of B scorzonerifolium could be achieved through multiple targets and components,providing a more reliable basis for the formulation and improvement of the quality standard of B.scorzonerifolium.
