δ-榄香烯体内外抗肿瘤作用研究 被引量:19

Anti-tumor Effects of δ-elemene in Vivo and in Vitro

摘要 目的:研究δ-榄香烯体内外抗肿瘤作用。方法:以肿瘤生长抑制率为指标,体外采用MTT法检测δ-榄香烯对人宫颈癌Hela、胃癌SGC-7901、结肠腺癌DLD-1、黏液性表皮样肺癌NCI-H292、乳腺癌MCF-7、肝癌HepG-2细胞及急性前髓细胞性白血病HL-60细胞和小鼠正常骨髓细胞及人正常肝胚胎细胞WRL-68的生长抑制作用;体内检测δ-榄香烯对裸鼠移植性人肝癌HepG2肿瘤细胞的生长抑制作用,电镜下观察细胞形态变化。结果:δ-榄香烯在体外对上述7种人肿瘤细胞的半数抑制浓度分别为157.9、323.6、222.4、233.2、287.3、231.6、154.5μmol·L-1,而对小鼠骨髓细胞和人正常肝胚胎细胞的生长无抑制。给药剂量为75mg·kg-1和150mg·kg-1的δ-榄香烯在体内对裸鼠移植性人肝癌HepG2细胞的生长抑制率分别为33.84%和47.47%;电镜下150mg·kg-1δ-榄香烯组细胞膜表面可见细胞表面泡状突起形成、微绒毛消失或基本消失、胞体固缩,并可见分离出的凋亡小体。结论:δ-榄香烯具有一定的抗肿瘤作用,此作用可能是通过诱导肿瘤细胞凋亡实现的。 OBJECTIVE: To study the antitumor effects of δ-elemene both in vivo and in vitro. METHODS: Using tumor growth inhibition rate as index, the in vitro effects of δ-elemene on human cervical cancer Hela cell, gastric cancer SGC- 7901 cell, colon cancer DLD 1 cell, mucinous epidermoid lung cancer NCl - H292 cell, breast cancer MCF- 7 cell, liver cancer HepG- 2 cell, acute mydoid leukemia HL- 60 cell and normal mouse bone marrow cell,human embryonic stem WRL- 68 cell were detected by MTT; and its in vivo anti - tumor effect on transplanted human HepG2 tumor cells in nude mice was detected as well, cell morphological changes observed by electron microscope.RESULTS: The half inhibitory concentration that δ- elemene on the abovementioned seven kinds of tumor cells were 157.9, 323.6, 222.4, 233.2,287.3, 231.6, 154.5μmol · L 1 but little inhibitory effect on normal mouse bone marrow celia and human embryonic stem cell. The growth inhi- bition rate that δ- elemene(75, 150 mg· kg^-1) on transplanted human HepG2 tumor cells in vivo were aa.s4% and 47.47%. electron microscope, δ-elemene 150 mg ·kg^-1 group cell surface membrane surface can be seen protruding bubble formation, microvilli disappearance or disappeared, cell body shrinkage, and can be seen isolated from the apoptotic bodies . CONCLUSION : The anti tumor effect of δ- elemene is possibly achieved by inducing apoptosis of tumor cells.
