P-gp/NF-kB协同作用的枸杞子-7防治子宫内膜异位症复发的作用机制分析

摘要 目的从多药耐药角度观察枸杞子-7防治子宫内膜异位症复发的作用机制。方法以手术法制备子宫内膜异位症复发大鼠模型,以枸杞子-7灌胃治疗14 d,分别在给药第1天、第7天和第14天测量异位包块大小,末次给药后麻醉处死大鼠,测定血清E2和P含量,观察子宫组织病理组织学变化,以免疫印迹法测定子宫组织NF-KB和P-gp水平。结果与空白组相比,模型组血清E2和P含量升高、NF-KB和P-gp表达升高;与模型组相比,枸杞子-7组血清E2和P含量下降,病理学变化好转,并子宫组织NF-KB和P-gp明显下降。结论调节子宫内膜异位症复发模型NF-KB和P-gp异常表达是枸杞子-7防治子宫内膜异位症复发可能的作用机制。 Objective To observe the mechanism of Fructus Lycii-7 preventing and treating the recurrence of endometriosis from the perspective of multidrug resistance.Methods The rat model of recurrent endometriosis was prepared by surgical and treated with Fructus Lycii-7 by gavage for 14 days.The ectopic parcel sizes were measured respectively on dosing day 1,day 7 and day 14.The rats were anesthetized and killed after the last administration.The contents of serum estradiol(E2) and progesterone(P) were measured, the histopathological changes of uterine tissue were observed, and the levels of nuclear transcription factor kB(NF-kB) and P-glycoprotein(P-gp) in uterine tissue were measured by Western blotting.Results Compared with those of the blank group, the contents of serum E2and P as well as the expressions of NF-kB and P-gp were increased in the model group.Compared with those of the model group, the contents of serum E2and P in Fructus Lycii-7 group were decreased, the pathological changes improved, and the expressions of NF-kB and P-gp in uterine tissue were decreased significantly.Conclusion Regulating the abnormal expressions of NF-kB and P-gp in endometriosis recurrence model is the possible mechanism of Fructus Lycii-7 preventing and treating endometriosis recurrence.
