摘要
目的探究miR-451、miR-133a-3p、miR-96表达与胃癌病理特征的相关性。方法取收治93例胃癌患者癌组织、癌旁组织标本进行免疫组化染色,对待测标本中miR-451、miR-133a-3p、miR-96表达进行检测,对比分析不同临床病理特征胃癌组织中miR-451、miR-133a-3p、miR-96表达情况,分析miR-451、miR-133a-3p、miR-96表达在胃癌组织中的相关性。结果胃癌患者癌组织中miR-451、miR-133a-3p表达水平低于癌旁组织,miR-96表达水平高于癌旁组织(P<0.05)。不同性别(男、女)、年龄(>60岁、≤60岁)患者癌组织中miR-451、miR-133a-3p、miR-96表达水平比较,差异无统计学意义(P>0.05)。有淋巴结转移患者miR-451、miR-133a-3p表达水平低于无淋巴结转移患者,miR-96表达水平高于无淋巴结转移患者(P<0.05);与黏膜层浸润患者相比,浆膜层浸润患者miR-451、miR-133a-3p表达水平较低,miR-96表达水平较高(P<0.05);与Ⅰ、Ⅱ期患者相比,Ⅲ、Ⅳ期胃癌患者miR-451、miR-133a-3p表达水平较低,miR-96表达水平较高(P<0.05);与高分化患者相比,中、低分化胃癌患者miR-451、miR-133a-3p表达水平较低,miR-96表达水平较高,与中分化患者相比,低分化胃癌患者miR-451、miR-133a-3p表达水平较低,miR-96表达水平较高(P<0.05)。胃癌组织中miR-451、miR-133a-3p表达水平呈正相关(r=0.262,P=0.011);miR-451、miR-96表达水平负相关(r=-0.228,P=0.028);miR-133a-3p、miR-96表达水平呈负相关(r=-0.301,P=0.003)。结论miR-451、miR-133a-3p、miR-96在胃癌中表达出现异常,且miR-451、miR-133a-3p、miR-96表达水平变化与胃癌患者淋巴结转移情况、浸润程度、TNM分期、分化程度相关,三者表达水平之间具有一定相关性,共同参与胃癌发展演进过程。
Objective To analyze the correlation between the expression of miR-451,miR-133 a-3 p and miR-96 and the pathological features of gastric cancer.Methods Immunohistochemistry staining of cancer tissues and adjacent noncancerous tissues from 93 patients with gastric cancer were performed to determine the expression of miR-451,miR-133 a-3 p,and miR-96.The expression profiles of miR-451,miR-133 a-3 p,and miR-96 in gastric cancer tissues with different clinicopathological characteristics were analyzed.Results miR-451 and miR-133 a-3 p expression was significantly lower and miR-96 expression was significantly higher in cancer tissues than that in adjacent noncancerous tissues(P<0.05).There was no significant difference in miR-451,miR-133 a-3 p,or miR-96 expression between cancer tissues of male and female,or between patients aged>60 years and those aged≤60 years(P>0.05).Patients with lymph node metastasis had significantly lower mir-451 and mir-133 a-3 p expression than patients without lymph node metastasis and higher miR-96 expression than patients without lymph node metastasis(P<0.05).Patients with serosal layer invasion had significantly lower miR-451 and miR-133 a-3 p expression compared with patients with mucosal layer invasion(P<0.05).Patients with Stage Ⅲ or Ⅳ gastric cancer had significantly lower miR-451 and miR-133 a-3 p expression,and significantly higher miR-96 expression compared with patients with Stage Ⅰ or Ⅱ gastric cancer(P<0.05).Patients with moderately and poor differentiated gastric cancer had significantly lower miR-451 and miR-133 a-3 p expression compared with patients with well and moderate differentiated disease(P<0.05).In gastric cancer,miR-451 and miR-133 a-3 p were positively correlated(r=0.262,P=0.011);miR-451 and miR-96 expression were negatively correlated(r=-0.228,P=0.028);and miR-133 a-3 p and miR-96 expression was negatively correlated(r=-0.301,P=0.003).Conclusion Aberrant expression of miR-451,miR-133 a-3 p,and miR-96 is observed in gastric cancer,correlated with lymph node metastasis status,extent of invasion,TNM stage,and degree of differentiation.They are correlated with each other and collectively involved in the process of gastric cancer development and evolution.
