基于高效液相色谱法测定饮料中新食品原料吡咯喹啉醌二钠的含量及质谱确证 被引量:2

Determination of New Food Ingredient Pyrroloquinoline Quinone Disodium in Beverages by High Performance Liquid Chromatography and Confirmation by Mass Spectrometry

摘要 通过优化样品前处理和色谱分析方法,建立了高效液相色谱测定饮料中新食品原料吡咯喹啉醌二钠的分析方法,并采用液相色谱串联三重四极杆质谱进行确证。在SB-C18(250 mm×4.6 mm,5μm)、色谱柱甲醇-0.01 mol/L乙酸铵(添加1.61 g四丁基溴化铵)为流动相、流速为1.0 mL/min及检测波长为250 nm的条件下对目标物进行分离;结果表明:目标化合物在1~100μg/mL有良好线性;检出限为3 mg/kg;定量限为10 mg/kg;在10~40 mg/kg,乳饮料(酸性乳饮料)中吡咯喹啉醌二钠的回收率为93.62%~100.8%,相对标准偏差(RSD)为0.61%~3.83%,果汁(FC果汁)中吡咯喹啉醌二钠的回收率为87.83%~100.2%,相对标准偏差(RSD)为1.01%~1.08%;该分析方法具有简便、准确、精密度高和回收率好等优点,适用于食品饮料中吡咯喹啉醌二钠含量的检测。 By optimizing the method of the sample pretreatment and analysis,a high performance liquid chromatography(HPLC)method was established for the determination of new food ingredient pyrroloquinoline quinone disodium in orange juice and milk beverage and is confirmed by liquid chromatography tandem triple quadrupole mass spectrometry.Separations were performed on a SBC18(250 mm×4.6 mm,5μm)column with methanol-0.01 mol/L ammonium acetate(added 1.61 g tetrabutylammonium bromide)as mobile phase,flow rate of 1.0 mL/min and measured wave length of 250 nm.Results indicated that pyrroloquinoline quinone disodium had good linear relation in the range of 1~100μg/mL;The detection limit was 3 mg/kg;The quantification limit was 10 mg/kg;at the content level of 10~40 mg/kg,the recovery of pyrrorolequinoline quinone disodium in milk drink(acid milk drink)was 93.62%~100.8%,the relative standard deviation method precision(RSD)was 0.61%~3.83%,the recovery of pyrroroquinoline quinone disodium in juice(FC juice)was 87.83%~100.2%,the relative standard deviation method precision(RSD)was 1.01%~1.08%;The analytical method has the advantages of simplicity,accuracy,precision and recovery,and is suitable for the detection of disodium pyrroloquinoline quinone in food and beverages.
