摘要
目的建立高效液相色谱法快速检测婴幼儿配方食品中维生素B_(12)的分析方法。方法选择C18柱(250mm×4.6mm,5.0μm)作为色谱柱,流动相为水+甲醇,流速0.5mL/min等度洗脱,进样量35μL,柱温40℃,波长361nm,样品经超声提取15min,后在8000r/min下冷冻离心10min,离心结束后取上清液过0.22μm滤膜,上机测试。结果维生素B12标准品在浓度0.05~2.0μg/mL范围内线性良好;相关系数r为0.9998;平均回收率为91.3%;检出限为2ng/mL。结论该方法重复性好、简单、快速、结果准确,可应用于婴幼儿配方食品中维生素B_(12)含量的检测。
Objective To establish a method for rapid detection of vitamin B_(12) in infant formula foods by high performance liquid chromatography.Methods C18 column(250 mm×4.6 mm,5.0μm)was selected as the chromatographic column,the mobile phase was water+methanol(25%),the flow rate was 0.5 mL/min using isocratic elution,the injection volume was 35μL,the column temperature was 40℃,and the wavelength was 361 nm.The sample was extracted by ultrasonic for 15 min,and then centrifuged at 8000 r/min for 10 min.After centrifugation,the supernatant was taken through a 0.22μm filter membrane and tested on the machine.Results The standard vitamin B_(12) had good linearity within the concentration range of 0.05-2.0μg/mL.The correlation coefficient r was 0.9998.The average recoveries was 91.3%,and the limit of detection was 2 ng/mL.Conclusion This method is repeatable,simple,rapid and accurate,whice can be applied to the determination of vitamin B_(12) in infant formula foods.
