摘要
目的探讨β-连环蛋白(β-catenin)、赖氨酰氧化酶样蛋白2(LOXL2)在骨肉瘤组织中表达的相关性及生物学意义。方法回顾性分析82例骨肉瘤患者资料,并以52例骨软骨瘤患者资料作为对照。采用Western blot法检测β-catenin、LOXL2的表达水平;比较β-catenin、LOXL2在不同临床病理特征骨肉瘤患者中的表达水平;比较不同β-catenin、LOXL2表达水平的骨肉瘤患者总生存情况;对骨肉瘤细胞株SAOS2中LOXL2靶向调控β-catenin的作用进行验证。结果①骨肉瘤组织中β-catenin、LOXL2的平均表达水平均明显高于骨软骨瘤组织(P<0.05),且骨肉瘤组织中β-catenin的表达水平与LOXL2的表达水平呈正相关。②随着肿瘤体积增大、Enneking分期增加、远处转移的发生,骨肉瘤组织中β-catenin、LOXL2的表达水平明显增加。③骨肉瘤患者总生存率β-catenin、LOXL2高表达组均明显低于低表达组(P<0.05)。④骨肉瘤细胞株SAOS2中LOXL2、β-catenin的表达水平及细胞增殖的A_(490 nm)水平、侵袭细胞的数目si-LOXL2组均明显低(少)于si-NC组(P<0.05)。结论骨肉瘤组织中β-catenin、LOXL2表达增加,与病理进展、预后不良有关;LOXL2在骨肉瘤组织中可能靶向调控β-catenin。
Objective To explore the correlation between the expression ofβ-catenin and lysine oxidase-like protein 2(LOXL2)in osteosarcoma and its biological significance.Methods The data of 82 patients with osteosarcoma were analyzed retrospectively,and data of 52 patients with osteochondroma were selected as control.The expression levels ofβ-catenin and LOXL2 were detected by Western blot method;the expression levels ofβ-catenin and LOXL2 among osteosarcoma patients with different clinicopathological features were compared;the overall survival situation role of osteosarcoma patients with different expression levels ofβ-catenin and LOXL2 were compared.Targeted regulation of LOXL2 onβ-catenin in osteosarcoma cell line SAOS2 were verified.Results①In osteosarcoma tissue,the average expression levels ofβ-catenin and LOXL2 were significantly higher than those in osteochondroma(P<0.05),and the expression level ofβ-catenin and were positively correlated with the expression level of LOXL2.②With the increase of tumor volume and Enneking stage,and distant metastasis occurrence,the expression levels ofβ-catenin and LOXL2 in osteosarcoma were significantly increased.③For overall survival rate of patients with osteosarcoma,β-catenin and LOXL2 in high expression group were significantly lower than those in low expression group(P<0.05).④In the cell line SAOS2 of osteosarcoma,the expression levels ofβ-catenin and LOXL2,cell proliferation A 490 nm level,invaded cell numbers of si-LOXL2 group were significantly lower(less)than those in si-NC group(P<0.05).Conclusions The expression increase ofβ-catenin and LOXL2 in osteosarcoma are related with pathological progress and poor prognosis.LOXL2 may target and regulateβ-catenin in osteosarcoma tissue.
