摘要
目的研究赖氨酰氧化酶样蛋白2(LOXL2)和I型胶原蛋白ɑ1链(COL1A1)在食管鳞癌中的表达,探索两者蛋白表达与患者临床病理特征的相关性。方法以2019年9月-12月新疆医科大学附属肿瘤医院收治的45例食管鳞癌患者为研究组,以同期3例健康体检者为对照组。采用实时定量聚合酶链反应(RT-PCR)方法检测LOXL2和COL1A1在食管鳞癌组织及癌旁组织中的表达水平;采用外泌体试剂盒提取血浆外泌体,免疫印迹试验(Western Blot)检测血浆外泌体LOXL2的表达。结果与癌旁组织相比,食管鳞癌组织中LOXL2 mRNA和COL1A1 mRNA相对表达量升高,差异有统计学意义(P<0.05)。LOXL2高表达与肿瘤分化程度(χ^(2)=8.879,P<0.05)、淋巴结转移(χ^(2)=4.549,P<0.05)、TNM分期(χ^(2)=7.123,P<0.05)具有相关性;COL1A1高表达与淋巴结转移(χ^(2)=4.874,P<0.05)、TNM分期(χ^(2)=8.186,P<0.05)有关。食管鳞癌组织中LOXL2和COL1A1表达呈正相关(r=0.7289,P<0.05)。与健康体检者比较,食管鳞癌患者的血浆外泌体LOXL2蛋白表达水平升高,差异有统计学意义(t=3.005,P<0.05)。结论LOXL2和COL1A1在食管鳞癌组织中高表达,并与区域淋巴结转移、临床分期相关,两者异常高表达与食管鳞癌的发生发展密切相关。
Objective To study the expression of lysine oxidase-like protein 2 (LOXL2) and type I collagen 1 chain(COL1A1) in esophageal squamous cell carcinoma tissues and explore the relationship between expression and clinicopathology. Methods The study group included 45 patients with esophageal squamous cell carcinoma admitted to the hospital from September to December 2019, and three healthy patients in the same period were taken as the control group.The expression level of LOXL2 and COL1A1 was measured by real-time and quantitative method (RT-PCR)plasma exosomes were extracted by exosome kit, and the expression of LOXL2 was measured by Western Blot. Results The relative expression of LOXL2 mRNA and COL1A1 mRNA in esophageal squamous carcinoma were compared with adjacent tissues (P<0.05). High expression of LOXL2 was significantly associated with the degree of tumor differentiation (χ^(2)=8.879,P<0.05), lymph node metastasis(χ^(2)=4.549,P<0.05) and TNM stage (χ^(2)=7.123,P<0.05);High expression of COL1A1 was associated with lymph node metastasis (χ^(2)=4.874, P<0.05), and TNM stage (χ^(2)=8.186, P<0.05). A positive correlation between LOXL2 and COL1A1 expression in esophageal squamous carcinoma tissues(r =0.7289, P<0.05). Plasma exosomal LOXL2 protein expression levels were increased in esophageal squamous carcinoma patients compared with healthy medical examinations (t=3.005, P<0.05). Conclusions LOXL2 and COL1A1 are highly expressed in esophageal squamous carcinoma tissues and are associated with regional lymph node metastasis clinical stage, and both of are closely related to development of esophageal squamous carcinoma.
