驴源枯草芽孢杆菌分离鉴定及生物学特性 被引量:5

摘要 为分离驴源具有抑菌活性的优良芽孢杆菌,本试验通过分离培养和形态观察,从健康驴新鲜粪便样品中分离芽孢杆菌,采用牛津杯法初步筛选具抑菌活性目标菌株,并进行生理生化和分子鉴定,研究其产酶活性、抑菌活性、耐酸性和耐胆盐性能、药物敏感性。结果显示:筛选到两株对金黄色葡萄球菌有强抑制作用的枯草芽孢杆菌(Bacillus subtilis),编号分别为LV1和LV2。两菌株具有很强的耐酸、耐胆盐特性,能产淀粉酶、纤维素酶和蛋白酶,并对检测用9种抗生素都敏感。结果表明,两株驴源枯草芽孢杆菌LV1和LV2具备良好的抑制金黄色葡萄球菌、产酶和耐酸、耐胆盐等益生活性,可作为研发驴用微生态制剂的潜在益生菌菌株。 The object of this work was to isolate Bacillus with antibacterial activity for research on probiotics of donkey.With method of isolation,culture and morphological observation,Bacillus was isolated from fresh feces and further screened based on antibacterial activity.Two isolates against Staphylococcus aureus were identified as Bacillus subtilis by physiological,biochemical characteristics and 16S rDNA sequencing,and termed LV1 and LV2.Biological properties such as enzyme producing activity,antibacterial activity,the resistance to acid condition and bile salt condition,and drug sensitivity were evaluated.The results showed that both LV1 and LV2 had good characteristics,including strong aintimicrobial activity against S.aureus,resistance to low pH and high bile salt concentration,production activity of cellulose,protease and amylase,and drug susceptibility to antibiotics used in this study.The results indicated that both B.subtilis strains from donkey performed better probiotic characteristics and could be used as a potential probiotic strains to develop probiotics for donkey.
