基于网络药理学探讨牛膝-钩藤药对治疗糖尿病合并高血压的作用机制 被引量:1

Discussion About the Mechanism of Action of Niuxi-Gouteng Drug Pair in the Treatment of Diabetes Complicated with Hypertension Based on Network Pharmacology

摘要 目的:基于网络药理学技术研究牛膝-钩藤药对治疗糖尿病合并高血压的作用机制。方法:基于中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)检索牛膝、钩藤的活性成分及相关作用靶点,利用Cytoscape 3.7.2软件构建“药物-活性成分-靶点”相互作用网络图;在人类孟德尔遗传数据库(online mendelian inheritance in man,OMIM)、人类基因数据库(GeneCards)、DrugBank数据库及疗效药靶数据库(therapeutic target database,TTD)中筛选糖尿病、高血压的相关靶点,与活性成分靶点进行映射,并绘制“活性成分-疾病”交集靶点韦恩图。利用STRING在线分析平台分析交集靶点的相互作用关系,利用CentiScaPe 2.2插件进行网络拓扑分析,筛选出牛膝-钩藤治疗糖尿病合并高血压的潜在核心靶点。采用基因注释和资源分析数据库(Metascape)对潜在核心靶点进行基因本体(gene ontology,GO)功能富集分析和京都基因与基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)通路富集分析,并利用Cytoscape 3.7.2软件构建“活性成分-核心靶点-通路-疾病”网络图。结果:共筛选出牛膝-钩藤活性成分46个及其作用靶点199个,糖尿病合并高血压相关靶点1 494个,将活性成分靶点与疾病靶点取交集,得到104个交集靶点。网络拓扑分析筛选出潜在核心靶点47个。GO富集分析得到生物过程1 004条、分子功能32条及细胞组成84条。KEGG通路富集分析得出153条信号通路。结论:牛膝-钩藤药对通过多成分、多靶点、多途径干预并治疗糖尿病合并高血压。 Objective:To study the mechanism of action of Niuxi(Radix Achyranthis Bidentatae)-Gouteng(Ramulus Uncariae Cum Uncis) drug pair in the treatment of diabetes complicated with hypertension based on network pharmacology technology.Methods:Based on the traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP),the effective components and related action targets of Niuxi and Gouteng were retrieved.The relevant targets of the active components of Niuxi-Gouteng were mapped with the active components by using Cytoscape 3.7.2 software to obtain the "drug-active component-target" interaction network diagram.In the online mendelian inheritance in man(OMIM),GeneCards,DrugBank and therapeutic target database(TTD),the targets related to diabetes and hypertension were screened,mapped with the targets of active ingredients,and the Wayne diagram of the intersection target of "active ingredients-disease" was drawn.By using the STRING online analysis platform,the interaction between intersecting targets was analyzed;By using the CentiScaPe 2.2 plug-in,the network topology parameters were carried out,and the potential core targets of Niuxi-Gouteng for the treatment of diabetes complicated with hypertension were screened.Gene ontology(GO) function enrichment analysis and Kyoto encyclopedia of genes and genomes(KEGG) pathway enrichment analysis were performed on the potential core targets of Niuxi-Gouteng for the treatment of diabetes complicated with hypertension by using the gene annotation and resource analysis database(Metascape),and the network diagram of "active ingredients-core targets-pathways-diseases" was constructed by using the Cytoscape 3.7.2 software.Results:A total of 46 active components of Niuxi-Gouteng and 199 action targets were screened,and 1,494 targets related to diabetes and hypertension were screened.A total of 104 intersection targets were obtained by taking the intersection of active component targets and disease targets.Through network topology analysis,47 potential core targets were screened.GO enrichment analysis yielded 1,004 biological processes,32 molecular functions and 84 cellular composition.The enrichment analysis of KEGG pathway yielded 153 signal pathways.Conclusion:Niuxi-Gouteng drug pair can intervene and treat diabetes complicated with hypertension through multi-component,multi-target and multi-channel.
