龙眼SPFH家族全基因组鉴定、进化分析与功能预测

摘要 SPFH(stomatin,prohibitin,flotillin and hflk/C)是一种膜微区蛋白,该家族的蛋白均含有一个高度保守的SPFH结构域.在植物中,SPFH家族蛋白可以细分为Slp(stomatin-like protein)、PHB(prohibitin)、Flot(flotillin)、Elp(erlin-like protein)以及HIR(hypersensitive induced reaction)5类.目前关于SPFH在植物中的全基因组鉴定鲜见报道,在拟南芥、水稻等植物中也只是对SPFH的个别亚家族进行研究.为了解膜微区蛋白SPFH在龙眼中的分子特性,基于龙眼基因组数据库,对龙眼SPFH(DlSPFH)进行家族成员鉴定、进化分析,并结合龙眼转录组数据库对DlSPFHs在早期体胚发生过程中的表达模式进行分析.结果显示,DlSPFH家族共有14个成员,包含DlSlp(1个)、DlElp(1个)、DlFlots(2个)、DlPHBs(2个)以及DlHIRs(8个)5个亚家族;家族成员的氨基酸数目、分子量、等电点分别介于193-2 570aa、21.49×10;-282.11×10;、5.38-9.42之间;定位于8条染色体上. DlSPFHs家族间存在2对串联重复基因以及3对片段重复基因.此外,DlSPFHs还与拟南芥、甜橙、水稻SPFH家族成员分别存在17、14、2对共线性成员.基因结构与保守基序分析结果显示,DlSPFH各成员间基因结构存在较大的差异;基序以motif 1最为保守.蛋白互作结果显示,DlSPFHs不仅家族成员之间存在互作关系,还能够与多种蛋白发生互作.DlSPFHs在启动子区域存在较多激素响应元件,并且可能受到11种miRNA的精确调控.在龙眼体胚发生早期阶段DlHIR1、DlHIR2、DlHIR3、DlPHB1、DlHIR8相较其他成员在体胚发生早期各个阶段呈现出较高的表达量FPKM值.本研究表明龙眼DlSPFHs的基因重复事件主要以片段重复为主,并且可能参与龙眼的抗病、激素响应以及体胚发生过程,展现功能多样性.(图7表3参39) A stomatin,prohibitin,flotillin,and hflk/C(SPFH)domian is a highly conserved membrane microdomain protein.In plants,the SPFH family of proteins comprises five subfamilies:stomatin-like protein(Slp),prohibitin(PHB),flotillin(Flot),Erlin-like protein(Elp),and hypersensitivity-induced reaction.There are few reports on the genome-wide identification of SPFH in plants,and only a few subfamilies of SPFH have been studied in plants,such as Arabidopsis thaliana and rice.To understand the molecular characteristics of membrane microdomain protein SPFH in longan,based on the longan genome database,family member identification and evolution analysis of longan SPFH(DlSPFH)was investigated.The data were combined with the longan transcriptome database to analyze the expression patterns of DlSPFHs during early somatic embryogenesis.The DlSPFH family contained 14 members,including five subfamilies:DlSlp(1),DlElp(1),DlFlots(2),DlPHBs(2),and DlHIRs(8).Features included 193-2570 amino acids,molecular weight ranging from 21.49×10^(3)-282.11×10^(3),and isoelectric point from 5.38-9.42.The 14 DISPFHs are located on eight chromosomes.Two pairs of tandem duplication genes and three pairs of fragment duplication genes were identified in the DlSPFH family.In addition,there were 17,14,and two pairs of collinearity members in the DlSPFHs and Arabidopsis,sweet orange,and rice SPFH family members,respectively.Analysis of the gene structure and conserved motifs revealed significant differences in gene structure among DlSPFH members.Motif 1 was the most conserved.Detection of protein showed that DlSPFHs interacted with family members and with a variety of proteins.DlSPFHs include many hormone response elements in the promoter region,which might be precisely regulated by 11 microRNAs.In the early stages of somatic embryogenesis,DlHIR1,DlHIR2,DlHIR3,DlPHB1,DlHIR8 showed higher expression levels of FPKM than other members.Gene duplication events of DlSPFHs were mainly fragment duplication and might be involved in disease resistance,hormone response,and somatic embryogenesis in longan.
