摘要
目的:探讨RNA结合蛋白构建的胃癌预后模型并验证其准确性。方法:从癌症基因组图谱(Cancer Genome Atlas,TCGA)数据库中下载胃癌的RNA测序数据,并测定正常组织和肿瘤组织中不同表达的RNA结合蛋白(RNA binding protein,RBPs)。并系统地研究及分析这些RBPs的表达和预后价值。结果:共鉴定296个不同表达的RBPs,其中166个表达上调,130个表达下调。7个RBPs(RNASE1、SETD7、BOLL、ADARB1、PPARGC1B、LARP6、MSI2)被鉴定为与预后相关的hub基因。基于以上基因构建的预后模型显示,高风险亚组患者总体生存期(OS)较低风险亚组患者低(P<0.05)。在train组中,预后模型的ROC曲线下的面积为0.730,test组中为0.653。列线图和HPA数据库进一步验证了胃癌和正常组织中7个关键RBPs的表达差异。结论:胃癌的RNA结合蛋白相关预后模型显示了良好的预后鉴别能力,它将有助于揭示胃癌的发病机制并提供新的临床诊疗思路。
Objective:To explore the prognostic model of gastric cancer with RNA binding protein and verify its accuracy by using bioinformatics.Methods:The RNA sequencing data of gastric cancer were downloaded from the Cancer Genome Atlas(TCGA)database,and the RBPs expressed in normal and tumor tissues were determined.The expression and prognostic value of these RBPs were studied systematically by bioinformatics analysis.Results:A total of 296 expressions of RBPs were identified,166 of which were up-regulated and 130 of which were down-regulated.Seven RBPs(RNASE1,SETD7,BOLL,ADARB1,PPARGC1B,LARP6,MSI2)were identified as prognostic hub genes.The prognostic model based on the above genes showed that the overall survival(OS)of the high-risk subgroup was lower than that of the low-risk subgroup.In the train group,the area under the ROC curve of the prognosis model was 0.730,while in the test group,it was 0.653.The expression differences of 7 RBPs in gastric cancer and normal tissue were further verified by the nomogram and HPA database.Conclusion:The RNA-binding protein-related prognostic model of gastric cancer shows a good prognostic differentiation ability,which will help to reveal the pathogenesis of gastric cancer and provide new clinical diagnosis and treatment ideas.
