摘要
目的探讨微小RNA-335(miR-335)在骨肉瘤患者血清中的表达及临床意义。方法选取42例骨肉瘤患者作为骨肉瘤组,42例健康志愿者作为对照组,采用实时荧光定量PCR检测两组血清miR-335水平并进行比较,分析血清miR-335水平与骨肉瘤患者临床特征的关系,采用受试者工作特征曲线评估血清miR-335对骨肉瘤的诊断价值。结果对照组血清miR-335水平为0.97±0.27,明显高于骨肉瘤组的血清miR-335水平(0.43±0.14),差异有统计学意义(t=11.53,P<0.05)。不同分化程度和有无远处转移的骨肉瘤患者血清miR-335水平比较,差异均有统计学意义(P<0.05)。血清miR-335诊断骨肉瘤的灵敏度为73.2%,特异度为87.8%,曲线下面积为0.824,截断值为0.55。结论骨肉瘤患者血清miR-335表达下调,其可能参与了骨肉瘤的发生、发展,并可作为骨肉瘤诊断标志物。
Objective To explore the expression and clinical significance of serum microRNA-335(miR-335) in patients with osteosarcoma.Methods Selected 42 patients with osteosarcoma as the osteosarcoma group and 42 healthy volunteers as the control group.Real-time fluorescence quantitative PCR was used to detect the serum miR-335 levels of the two groups and compared.Analyzed the relationship between serum miR-335 level and clinical characteristics of patients with osteosarcoma.The receiver operating characteristic curve was used to evaluate the diagnostic value of serum miR-335 for osteosarcoma.Results The serum miR-335 level of control group was 0.97±0.27,which was significantly higher than that of osteosarcoma group(0.43±0.14),and the difference was statistically significant(t=11.53,P<0.05).There were statistically significant differences on serum miR-335 levels of osteosarcoma patients with different degrees of differentiation and with or without distant metastasis(P<0.05).The sensitivity of serum miR-335 in diagnosing osteosarcoma was 73.2%,the specificity was 87.8%,the area under the curve was 0.824,and the cut-off value was 0.55.Conclusion The expression of serum miR-335 is down-regulated in patients with osteosarcoma, which may be involved in the occurrence and development of osteosarcoma and can be used as a marker for diagnosis of osteosarcoma.
