摘要
研究了梵净山4种不同叶片表型的紫苏籽(PFS)油的提取及其脂肪酸组成。在单因素试验基础上通过响应面法优化得到最优超声辅助提取条件为以石油醚为溶剂,超声频率40 k Hz、功率300 W、提取1次、时间10 min、提取温度35℃、料液比1∶18(g/m L)。利用气质联用技术(gas chromatography-mass spectrometry,GC-MS)测定4种不同叶型的紫苏籽油的脂肪酸组成。PFS1、PFS2、PFS3、PFS4的提油率分别为(36.63±0.75)%、(34.89±0.66)%、(36.44±0.67)%、(36.68±0.37)%,α-亚麻酸含量分别为54.45%、50.42%、67.04%、69.81%;不饱和脂肪酸(PUFAs)的相对含量表现出与α-亚麻酸变化相同的规律,其中PFS3、PFS4的PUFAs相对含量分别为91.81%、92.34%。结果表明梵净山紫苏籽(PFS3、PFS4)是天然植物保健油优质资源,同时提示紫苏籽油含量及脂肪酸组成与紫苏叶型存在相关性。
The extraction and fatty acid composition of four different leaf phenotypes of Perilla seed(PFS)oil from Fanjing Mountain were studied.Based on the single factor test results,process of ultrasound-assisted extraction of the oils of Perilla frutescen seed(PFS)was optimized by response surface method(RSM).The research showed that the optimum parameters was extraction time 10 min,extraction temperature35℃,material-liquid ratio 1∶18(g/m L),times of 1,frequency 40 k Hz,power 300 W using petroleum ether as the extraction solvent.The fatty acids composition was determined by gas chromatography-mass spectrometry(GC-MS).The oil extraction rates of FPS1,FPS2,FPS3,FPS4 were(36.63±0.75)%,(34.89±0.66)%,(36.44±0.67)%,(36.68±0.37)%,respectively.And the relative contents ofα-linotenic acid in these samples from FPS1 or FPS2 or FPS3 or FPS4 were 54.45%,50.42%,67.04%,69.81%,respectively.The relative content of unsaturated fatty acids(PUFAs)was the same as that ofα-linolenic acid,and the relative content of PUFAs in PFS3 and PFS4 was91.81%and 92.34%,respectively.The results showed PFS3 or PFS4 from Fanjing Mountain was a good resource of natural health care oil.Simultaneously,it was indicated that there was a correlation between leaf morphology and the content of Perilla seed oil andα-linolenic acid.
