摘要
目的:建立HPLC法测定10种潮州凤凰茶叶中咖啡因的含量。方法:以Agilent ZORBAXEclipse XDB-C18为分析柱,样品进样体积为10μL。流动相A为0.2%的乙腈溶液,流动相B为55%甲醇,流速为1ml/min,柱温箱为30℃,紫外检测器检测波长272nm。结果:在所选条件下,样品中咖啡因与其它组分分离良好。在咖啡因浓度为0.02~0.16mg/mL范围内与峰面积有良好的线性关系,相关线性系数达到0.99999,加标回收率97.75%~99.83%之间,平均回收率为98.65%,RSD=0.8177%。10种凤凰单丛茶咖啡因含量在20.50-31.00mg/g之间。结论:该方法快速准确,可用于茶叶中咖啡因含量的测定。
AIM: To establish an HPLC method for the content determination of caffeine in ten kinds of Fenghuang tea from Chaozhou. METHODS: We taked Agilent ZORBAXEclipse XDB-C18 as analytical column, the inject volume was 10μL, and taked 0.2% acetonitrile solution as the mobile phase A and 55% methanol solution as the mobile phase B, flow rate was 1ml/min, the column temperature was 30℃,and the detection wavelength was 272nm.Under the selected conditions. RESULTS: The caffeine got an ideal separation with other compounds. The peak time of caf⁃ feine has a good liner relation with the peak area of caffeine when the concentration of caffeine range of 0.02 to 0.16mg/mL, and the relevant linear coefficient reached 0.99999(n= 5). In addition, the adding standard recoveries were 97.75%~99.83%, the average recoveries were 98.65%, and RSD=0.8177% .The caffeine content of ten kinds of Fenghuang Dancong tea ranged from 20.50 to 31.00mg/g.CONCLUSION: This method was quick and accurate. Therefor it was suitable for the determination of caffeine in tea.
