茶多酚的指纹图谱和主要成分的含量测定方法研究 被引量:14

摘要 目的:建立茶多酚的 HPLC 指纹图谱检测方法。方法:采用 ODS 柱(250 mm×4.6 mm,5μm);流动相 A:甲醇-0.1%甲酸溶液(10:90),流动相 B:甲醇-乙腈-0.1%甲酸溶液(50:30:20),进行梯度洗脱:0 min(0%B)→10 min(10%B)→30min(20%B)→40 min(30%B)→45 min(40%B)→55 min(50%B);流速1 mL·min^(-1);检测波长280 nm。并对主要成分峰进行 LC-MS/MS 鉴定。结果:茶多酚 HPLC 指纹图谱中有11个特征成分峰,经对照品对照及液质联用鉴定了其中9个成分,分别为表儿茶素(EC)、儿茶素(C)、表没食子儿茶素(EGC)、没食子儿茶素(GC)、表儿茶素没食子酸酯(ECG)、儿茶素没食子酸酯(CG)、表没食子儿茶素没食子酸酯(EGCG)、没食子儿茶素没食子酸酯(GCG)和咖啡因(CAF)。结论:HPLC 指纹图谱可以对茶多酚中主要特征有效成分进行准确的定性与定量测定,色谱系统稳定,适用性良好。 Objective:To establish the fingerprint chromatogram standard of tea polyphenols by HPLC. Method: Separation was performed on ODS column(250 mm×4. 6 mm ,5μm)with gradient elution by the mobile phase consisting of methanol - 0. 1% formic acid ( 10: 90) and methanol - acetonitrile - 0. 1% formic acid ( 50: 30: 20 ). The detection wavelength was set at 280 nm. And most of chromatogram peaks were identified by LC - MS/MS. Result: There were 11 common peaks in fingerprint chromatograms and 9 of them were identified with reference substances and LC - MS/MS, which were ( - ) - epicatechin ( EC ), ( + ) - catechin ( C ), ( - ) - epigallocatechin ( EGC ), ( - ) - gallocatechin (GC), ( - ) - epigallocatechin gallate ( EGCG), ( - ) - gallocatechin gallate (GCG), ( - ) - epicatechingallate ( ECG), ( - ) - catechingallate ( CG), caffeine ( CAF), and etc. Conclusion: The method is accurate in identification and determination of compounds in tea polyphenols. The chromatographic system is stable and applicable.
