摘要
为探究不同党参炮制四君子汤(SJZD)对脾虚家兔治疗作用及机理,将50只健康家兔随机分为对照组、模型组、SJZD-Ⅰ组、SJZD-Ⅱ组和SJZD-Ⅲ组,每组10只;除对照组外,其余各组家兔用灌胃厚朴三物汤加饥饱失常法造脾虚模型。SJZD-Ⅰ、SJZD-Ⅱ和SJZD-Ⅲ组脾虚家兔分别用含党参、米党参和蜜党参的四君子汤治疗,采用ELISA法测定家兔血清中胃肠激素(GAS、MTL)、IL-2、IL-4、IFN-γ、cAMP和cGMP水平。结果显示,SJZD-Ⅰ、SJZD-Ⅱ和SJZD-Ⅲ组脾虚家兔的治愈率分别达到80%,80%和90%。与对照组相比,模型组家兔血清中GAS、MTL水平显著降低(P<0.05),IL-2、IFN-γ、cAMP水平及cAMP/cGMP比值极显著降低(P<0.01),IL-4水平显著升高(P<0.05),cGMP水平极显著升高(P<0.01)。与模型组相比,SJZD-Ⅰ、SJZD-Ⅱ、SJZD-Ⅲ组家兔血清中GAS水平显著升高(P<0.05),MTL、IL-2、IFN-γ、cAMP水平及cAMP/cGMP比值极显著升高(P<0.01),IL-4和cGMP水平有降低的趋势;其中SJZD-Ⅱ组家兔血清中IL-4和cGMP水平显著或极显著降低(P<0.05或P<0.01),SJZD-Ⅲ组家兔血清中cGMP水平显著降低(P<0.05)。综上所述,不同党参炮制四君子汤均能有效治疗家兔脾虚证,可能与调节胃肠激素(GAS、MTL)、免疫因子(IL-2、IFN-γ、IL-4)及cAMP/cGMP分泌紊乱有关。
To investigate the therapeutic effects and mechanism of Sijunzi decoction(SJZD)with different processed Codonopsis on spleen deficiency in rabbits.Fifty healthy rabbits were randomly divided into control group,model group,SJZD-Ⅰgroup,SJZD-Ⅱgroup and SJZD-Ⅲgroup,with 10rabbits in each group.Except the control group,each group was fed with Houpu Sanwu decoction plus hunger and stagnation to establish a model of spleen deficiency syndrome in rabbits.SJZD-Ⅰgroup,SJZD-Ⅱgroup and SJZD-Ⅲgroup were treated by Sijunzi decoction with Codonopsis,Sijunzi decoction with rice processed Codonopsis and Sijunzi decoction with honey processed Codonopsis,respectively.The level of GAS,MTL,IL-2,IL-4,IFN-γ,cAMP and cGMP in the serum of rabbits were determined by ELISA.The results showed that the cure rate of spleen deficiency in rabbits in SJZD-Ⅰ,SJZD-Ⅱ,SJZD-Ⅲgroup were 80%,80%,90%,respectively.The levels of GAS,MTL,IL-2,IFN-γ,cAMP,cAMP/cGMP in serum of model group were significantly decreased(P<0.01or P<0.05),and the levels of IL-4,cGMP in serum of model group were increased significantly(P<0.01or P<0.05)in comparison to the control group.Compared with the model group,the serum level of GAS,MTL,IL-2,IFN-γ,cAMP,cAMP/cGMP in SJZD-Ⅰ,SJZD-Ⅱ,SJZD-Ⅲgroup were notably enhanced(P<0.01),and the serum level of IL-4,cGMP in SJZD-Ⅰ,SJZD-Ⅱ,SJZD-Ⅲgroup had a downward trend,Thereinto,the serum level of IL-4,cGMP in SJZD-Ⅱgroup were significantly reduced(P<0.01or P<0.05),and the serum level of cGMP in SJZD-Ⅲgroup was extremely decreased(P<0.01).In conclusion,Sijunzi decoction with different processed Codonopsis can effectively treat the spleen deficiency of rabbits,which may be related to the regulation of secretion disorder of gastrointestinal hormones(GAS,MTL),immune factors(IL-2,IFN-γ,IL-4)and cAMP/cGMP.
