摘要
目的建立基于一测多评法(QAMS)测定小儿解感颗粒中黄芩苷、汉黄芩苷、黄芩素、汉黄芩素、甘草苷5个成分的含量的方法。方法采用高效液相色谱(HPLC),色谱柱为Waters SunFire C18柱(4.6 mm×250 mm,5μm);流动相为乙腈(A)-0.1%磷酸水溶液(B),梯度洗脱;流速1.0 ml/min;柱温30℃;检测波长274 nm。以黄芩苷为内参物,建立汉黄芩苷、黄芩素、汉黄芩素、甘草苷与内参物黄芩苷的相对校正因子(f),通过f计算小儿解感颗粒中5个成分的含量,并进行验证,将该方法与外标法的测定结果进行对比分析,以验证QAMS方法的准确性、重复性及可行性。结果黄芩苷、汉黄芩苷、黄芩素、汉黄芩素、甘草苷质量浓度分别在0.102~407.492μg/ml、0.067~84.147μg/ml、0.154~60.197μg/ml、0.054~50.634μg/ml、0.272~237.796μg/ml范围内线性关系良好;加样回收率分别为99.16%,100.54%,98.02%,97.84%,98.78%;以黄芩苷为内参物,测得的汉黄芩苷、甘草苷、黄芩素、汉黄芩素f值分别为0.823,0.674,0.549,1.873,且建立的QAMS法结果与外标法的结果一致。结论本实验建立的QAMS法可作为一种简便快捷的质量评价模式,用于小儿解感颗粒中5种成分的质量控制。
Objective To establish a method for simultaneous determination of 5 components including baicalin,wogonoside,baicalein,wogonin and liquiritin in Xiao’er Jiegan Granules based on quantitative analysis of multicomponents with single-marker(QAMS).Methods The HPLC separation was achieved on a Waters SunFire C18 column(4.6 mm×250 mm,5μm)with acetonitrile(A)-0.1%phosphoric acid(B)as mobile phase of gradient elution,which was at a flow rate of 1.0 ml/min.The column temperature was set at 30℃.The determination wavelength was 274 nm.With baicalin as the internal reference standard,the relative correction factor(f)of wogonoside,baicalein,wogonin and liquiritin were calculated.The method was evaluated by comparison of the results between external standard method(ESM)and QAMS.Results Baicalin,wogonoside,baicalein,wogonin and liquiritin had good linear relations within the range of 0.102-407.492μg/ml,0.067-84.147μg/ml,0.154-60.197μg/ml,0.054-50.634μg/ml and 0.272-237.796μg/ml,respectively.The recoveries of the samples were 99.16%,100.54%,98.02%,97.84%,98.78%,respectively.With baicalin as the internal reference standard,the relative correction factors(f)were 0.823,0.674,0.549 and 1.873,respectively.There was no significant difference between the results of QAMS and ESM.Conclusion The QAMS method is simple and fast,and can be used to determine the multiple components in Xiao’er Jiegan Granules.
