摘要
为了获得药用价值较高的桑树原料,以我国北方地区的3个耐寒桑品种国桑21号、冀桑2号和桲椤为试材,采用氯甲酸芴甲酯(FMOC-Cl)柱前衍生RP-HPLC荧光检测法,对桑树不同不同部位以及不同叶位桑叶和叶片不同部位的1-脱氧野尻霉素(1-deoxynojirimycin,DNJ)含量进行了测定,并比较了DNJ含量在不同部位、不同叶位间的变化规律。结果表明:同一品种不同叶位桑叶的DNJ含量均随叶位(成熟度)的增加而逐渐降低,且桲椤的DNJ含量显著高于国桑21号和冀桑2号;桲椤和冀桑2号桑枝皮层的DNJ含量均明显高于桑叶,其中桲椤皮层中的含量显著高于冀桑2号,桑枝不同部位的皮层中DNJ含量顺序为上部<中部<下部;冀桑2号桑叶不同部位的DNJ含量顺序为叶片>叶柄>全叶>叶脉,但差异均不显著,其中叶片和叶脉的上半部DNJ含量高于下半部。河北省传统抗寒桑树品种桲椤桑的DNJ综合含量较高,其中皮层含量远高于桑叶。桑树中DNJ含量与品种、叶位和采摘部位等密切相关,DNJ的合成可能与叶片的生长有关,DNJ合成后通过叶脉和叶柄进而运输到桑树的各个部位。
To obtain mulberry material with high medicinal value,using three cold-resistant resources cultivated in east China as the test materials,the contents of 1-deoxyno-jirimycin(DNJ)in different parts of trees,different leaf positions and different parts of leaves were determined by derivatization with 9-fluorenylmethyl chloroformate(FMOC-Cl)and followed by reversed-phase high-performance liquid chromatography(RP-HPLC)and a fluorescence detector,whose variation regularity in different parts were also compared.The results showed that the contents of DNJ in different leaf positions gradually decreased with the increase of leaf positions(maturity),those of Poluo was significantly higher than those of Guosang No.21 and Jisang No.2.The contents in barks were obviously higher than those in leaves of Poluo and Jisang No.2,The content in barks of Poluo was significant higher than Jisang No.2.The order of DNJ content in barks was upper part<middle part<lower part.There was no significant difference in the content of DNJ in different parts of leaves,the order was leaves>petioles>whole leaves>leaves vein,and the content in the lower part was higher than that in the upper part of leaves and leaves vein.As a traditional cold-resistant mulberry variety in Hebei,Poluo has higher DNJ content,and the content in bark is much larger than in leaves.The contents of DNJ were correlated with mulberry variety,leaf position and collection part.The biosynthesis of DNJ may be related to the growth of leaves,and then transported to various parts of trees through veins and petioles.
