复合免疫亲和柱净化高效液相色谱法同时测定小麦中的4种真菌毒素 被引量:9

Simultaneous Determination of Four Mycotoxins in Wheat by High Performance Liquid Chromatography with Combination Immunoaffinity Column Purification

摘要 建立了一种复合免疫亲和柱净化高效液相色谱法同时测定小麦中脱氧雪腐镰刀菌烯醇、玉米赤霉烯酮、黄曲霉毒素B1和赭曲霉毒素A 4种真菌毒素的分析方法。样品利用80%的乙腈水溶液(v/v)提取后,采用复合免疫亲和柱净化,并通过高效液相色谱仪进行分离。色谱柱为ZORBAX Eclipse Plus C18柱(250 mm×4.6 mm,5μm),流动相A为0.1%的甲酸水溶液(v/v),流动相B为乙腈,梯度洗脱,流速1.0 mL/min,进样量50μL,光化学衍生器柱后衍生,二极管阵列检测器和荧光检测器同时检测。结果表明,样品在20 min内可以完成4种真菌毒素的液相分析,标准质控样检测偏差为—10%~+10%,在相应的线性范围内,回归方程的相关系数均超过0.999,加标回收率为86.7%~101.1%,精密度均小于5.0%。该方法简便、快速、准确、高效,适用于小麦中4种真菌毒素的同时快速测定。 A method for simultaneous determination of deoxynivalenol(DON),zearalenone(ZEN),aflatoxin B1(AFB1)and ochratoxin A(OTA)in wheat by high performance liquid chromatography(HPLC)with combination immunoaffinity column purification was developed.The mycotoxins were extracted by 80%(v/v)acetonitrile aqueous solution,purified by combination immunoaffinity column and separated by HPLC with diode array detector and fluorescence detector after post-column photochemical derivatization.The separation was performed on a ZORBAX Eclipse Plus C18 column(250 mm×4.6 mm,5μm).The mobile phase consisted of 0.1%(v/v)formic acid aqueous solution(A)and acetonitrile(B)by gradient elution,and the flow rate was 1.0 mL/min.The injection volume was 50μL.The results showed that the detection of four mycotoxins was completed within 20 minutes.The detection deviations of standard quality control sample were in the range of-10%~+10%.In the linear ranges,the correlation coefficients of regression equations were all over 0.999,the recoveries of standard addition were 86.7%~101.1%,and the relative standard deviations(RSDs)of precision were all less than 5.0%.The method is simple,rapid,accurate and efficient,and is suitable for the simultaneous and rapid determination of four mycotoxins in wheat.
