摘要
目的探讨术后血清细胞外基质蛋白转化生长因子β诱导(TGFBI)对胃癌复发转移的影响及其作用机制。方法利用shRNA结合慢病毒技术稳定敲低胃癌细胞SGC-7901中TGFBI的表达,并通过Western blot对其在细胞内及培养上清中的表达进行验证;MTT法检测TGFBI下调对胃癌细胞SGC-7901增殖的影响,Transwell法检测其对细胞侵袭与迁移的影响;通过检测E-cadherin、Snail和ZEB1等转录因子的表达,探究TGFBI是否通过上皮间质转化促进胃癌的转移复发。结果胃癌患者血清样本中TGFBI表达水平显著高于正常样本,RNA干扰后TGFBI在胃癌细胞SGC-7901内及其培养上清中的表达水平均发生显著下降;TGFBI下调后胃癌细胞SGC-7901的增殖、侵袭与迁移能力均受到显著抑制,同时上皮间质转化标志物E-cadherin的表达发生上调,而Snail和ZEB1的表达发生下调。结论胃癌细胞SGC-7901中TGFBI表达下调时可能通过抑制上皮间质转化,抑制胃癌细胞增殖、侵袭与迁移,最终抑制胃癌的转移复发。
Objective The aim of this study was to address the relevance of extracellular matrix transforming growth factorβ(TGFBI)to gastric cancer recurrence and metastasis and its probable mechanism.Methods TGFBI in gastric cancer cell SGC-7901 was stably down-regulated by shRNA combing with lentivirus technology and its expression level intracellular and in serum were detected by Western blot.MTT was conducted to investigate the influence of lowering TGFBI expression on gastric cancer cell SGC-7901 proliferation and Transwell was carried out to evaluate the cell invasion and migration under its action.Besides,expression level of transcription factor E-cadherin,Snail and ZEB1 were executed to investigate whether TGFBI promoted the metastasis and recurrence of gastric cancer by epithelial mesenchymal transition.Results TGFBI expression level in serum of gastric cancer patients was significantly higher than the normal samples,yet fell off in the gastric cancer cell SGC-7901 and its culture supernatant after RNA interfering.TGFBI down-regulation caused a notably decrease of gastric cancer cell SGC-7901 in its proliferation,invasion and migration.Synchronously,expression level of epithelial mesenchymal transition marker E-cadherin was up-regulated while the Snail and ZEB1 were down-regulated.Conclusion TGFBI down-regulation in gastric cancer cell SGC-7901 might mediated the restrain of gastric cancer cell proliferation,invasion and migration by epithelial mesenchymal transition,which ultimately inhibited the metastasis and recurrence of gastric cancer.
