摘要
分析川崎病患儿外周血单个核细胞单核细胞趋化蛋白1(MCP-1)mRNA表达水平及其临床意义。筛选我院2018年6月~2019年6月收治的70例川崎病患儿(急性期35例、亚急性期25例、恢复期10例)为川崎病组,另选同期70例健康体检儿童为对照组,采集空腹静脉血3 m L后利用酶联免疫吸附测定法检测血清MCP-1,荧光定量聚合酶链式反应(QF-PCR)测定单个核细胞MCP-1 mRNA表达水平,对两组以及川崎病组内不同临床分期患儿检测结果进行对比,以Pearson相关性分析法检验外周血单个核细胞MCP-1 mRNA表达水平与川崎病患儿临床分期的相关性。对照组血清MCP-1(45.45±2.35)pg/m L、外周血单个核细胞MCP-1 mRNA相对表达量(0.48±0.12),低于川崎病组(351.85±2.48)pg/m L、(1.72±0.18),差异有统计学意义(P<0.05);川崎病组内急性期患儿、亚急性期患儿、恢复期患儿血清MCP-1、外周血单个核细胞MCP-1 mRNA相对表达量分别为(514.88±3.60)pg/m L、(351.92±3.33)pg/m L、(189.78±3.42)pg/m L和(2.32±0.15)、(1.73±0.17)、(1.12±0.19),急性期患儿最高,恢复期患儿最低,亚急性期患儿介于其中,差异有统计学意义(P<0.05);Pearson相关性分析结果提示,外周血单个核细胞MCP-1 mRNA表达水平与川崎病临床分期呈正相关性(r=0.74,P<0.05)。川崎病患儿外周血单个核细胞MCP-1 mRNA表达水平与临床分期呈正相关性,对其数值进行检测有助于预测川崎病的发生与否,并为判断临床分期提供帮助。
To analyze the expression level and clinical significance of monocyte chemoattractant protein 1(MCP-1) mRNA in peripheral blood mononuclear cells of children with Kawasaki disease,Screening 70 children with Kawasaki disease(35 in the acute phase,25 in the subacute phase,and 10 in the recovery phase) admitted to our hospital from June 2018 to June 2019 were selected as the Kawasaki disease group.70 healthy children were used as the control group.After collecting 3 m L of fasting venous blood,serum MCP-1 was detected by enzyme-linked immunosorbent assay.Fluorescence quantitative polymerase chain reaction(QF-PCR)was used to determine the expression level of MCP-1 mRNA in mononuclear cells.In addition,the test results of children with different clinical stages in the Kawasaki disease group were compared.The correlation between the expression level of MCP-1 mRNA in peripheral blood mononuclear cells and the clinical stage of children with Kawasaki disease was tested by Pearson correlation analysis.The serum MCP-1(45.45 ± 2.35) pg/m L and the relative expression of MCP-1 mRNA in peripheral blood mononuclear cells(0.48 ± 0.12) were lower than those in Kawasaki disease group(351.85 ± 2.48) pg/m L,(1.72 ± 0.18),the difference was statistically significant(P<0.05).The relative expression of MCP-1 mRNA in serum MCP-1 and peripheral blood mononuclear cells in children with acute phase,subacute phase and convalescent phase in Kawasaki disease group were(514.88 ± 3.60) pg/m L,(351.92 ± 3.33) pg/m L,(189.78 ± 3.42) pg/m L and(2.32 ± 0.15),(1.73 ± 0.17),(1.12 ± 0.19),respectively.The children of acute phase were the highest,the children in the recovery period were the lowest,and the children in the subacute phase were among them,the difference was statistically significant(P<0.05).The results of Pearson correlation analysis indicated that the expression level of MCP-1 mRNA in peripheral blood mononuclear cells was different from that of Kawasaki.The clinical stage of the disease was positively correlated(r = 0.74,P<0.05).The expression of MCP-1 mRNA in peripheral blood mononuclear cells of Kawasaki disease is positively correlated with clinical stage.The detection of its value can help predict the occurrence of Kawasaki disease and help to determine clinical stage.
