摘要 目的:采用多指标综合评分法结合响应面法优选紫花前胡香豆素类成分的最佳提取工艺参数。方法:通过HPLC检测紫花前胡中的化学成分伞形花内酯、紫花前胡苷、补骨脂素、花椒毒素和佛手柑内酯的含量,以综合评分值作为评价指标,并对液料比、甲醇浓度和超声时间进行考察,且在单因素试验基础上,运用Box-Behnken设计响应面法优化提取工艺,并进行验证试验。结果:最佳提取工艺:液料比33.69 mL·g-1,甲醇浓度80.32%,超声时间46.64 min,在此条件下5种香豆素类成分提取综合评分值为5.9401,验证试验结果与预测值一致。结论:响应面法结合综合评分法优选出5个香豆素类成分的提取工艺稳定可靠,建立的提取方法简单可行,可对进一步研究紫花前胡有效成分开发利用提供参考。 Objective:To optimize the extraction on process of coumarins in Peucedanum decursivum Maxim by comprehensive score combined with Box-Behnken response surface methology.Methods:The HPLC was used to detect umbelliferone,nodakenin,psoralen,xanthotoxin and bergapten with the comprehensive score value as the evaluation index,and the liquid-solid ration,methanol concentration and extraction time were investigated.On the basis of the single-factor test,Box-Behnken design-response surface method was used to optimize the extraction process,and validation test was also conducted.Results:Optimal extraction process was as following:liquid-solid ration was 33.69 mL·g-1,methanol concentration was 80.32%,extraction time was 46.64 min,the five coumarins comprehensive score was 5.9401.The verification test results were consistent with the predicted values.Conclusion:Optimization of extraction process parameters of five coumarins in P.decursivum Maxim by Box-Behnken response surface methology with comprehensive scoring method.The established extraction method was simple,feasible,stable and reliable.The extract of P.decursivum Maxim can provide references for further study on the extraction of effective components from P.decursivum Maxim.
