基于体外抗氧化活性的山楂醇沉工艺优化 被引量:1

Optimization of the ethanol precipitation of Crataegus pinnatifida fruit based on antioxidant activity in vitro

摘要 目的确定山楂最佳醇沉工艺。方法利用Plackett-Burman实验设计法考察不同工艺参数对固体保留率(RDM)、总酚酸保留率(RTP)及反映DPPH自由基清除能力IC50的影响,进而采用Box-Behnken响应面法对关键工艺参数优化。结果山楂最佳醇沉工参数为:搅拌速度250 r·min-1,醇料比1.70 mL·g-1,醇沉前浓缩液密度为1.08g·mL-1。在此条件下,RDM、RTP及IC50分别为59.44%、72.61%及1.62mg·mL-1。结论采用Plackett-Burman实验设计法结合Box-Behnken响应面法优化山楂醇沉工艺简单可行,可预测性好。 Objective To optimize the ethanol precipitation process of Crataegus pinnatifida fruit.Methods Active ingredients from Crataegus pinnatifida fruit were extracted by water extraction and ethanol precipitation method,with the recovery of dry matter(RDM),the recovery of the total phenolic content(RTP)and IC50 value as index,the key process parameters were firstly evaluated by Plackett-Burman design.Then,the key process parameters were optimized by Box-Behnken design,multivariate linear regression and binomial fitting were applied to the experimental data,and the response surface method was used to predict and analyze the optimal process.Results The final optimized parameters were determined to be a stirring speed of 250 r·min-1,ethanol to material ratio of 1.70 mL·g-1,and extract density of 1.08 g·ml-1.Under the optimized conditions,the RDM,RTP and the IC50 were 59.44%,72.61%and 1.62 mg·mL-1,respectively.These results were in good agreement with the predicated results.Conclusion This ethanol precipitate on process is reasonable and feasible by Plackett-Burman design and response surface analysis with good predict ability,which provides support for the effective utilization of Crataegus pinnatifida fruit.
