富血小板纤维蛋白对兔耳软骨移植成活情况的影响 被引量:1

摘要 目的:研究自体富血小板纤维蛋白(Platelet-rich fibrin,PRF)对兔耳软骨移植后成活的影响,为临床应用提供证据。方法:选取16只新西兰大白兔,于兔双耳对称处设计并剥离出大小、形状相似,重量为80mg的耳软骨块,双侧耳软骨块随机分为对照组与实验组,对照组耳软骨块直接置入兔背部左侧浅筋膜腔隙内,实验组耳软骨块联合PRF共同置入兔背部右侧浅筋膜腔隙内。分别于2周、4周、8周、12周取材进行大体观察、重量比较及统计学分析、染色观察(HE染色、番红〇染色、阿利新蓝染色)。结果:通过大体观察和软骨重量可见:2~4周耳软骨块重量增加,8~12周耳软骨块重量减小,各时间点实验组耳软骨块重量均大于对照组,两组间比较差异有统计学意义(P<0.05)。染色观察可见:各时间点在相同显微倍数、同一视野下实验组软骨细胞明显多于对照组,其中新生软骨细胞也多于对照组。实验组异染性软骨基质、蓝色的糖胺聚糖均更明显。结论:自体富血小板纤维蛋白能够促进兔耳软骨移植后的存活。 Objective To study the effect of autologous platelet-rich fibrin(PRF)on the survival of rabbit ear cartilage after transplantation,so as to provide evidence for clinical application.Methods Sixteen New Zealand white rabbits were selected to design and strip out ear cartilage with similar size and shape and weight of 80mg on the symmetry of both ears.Both sides of ear cartilages were randomly divided into the control group and the experimental group.The piece of ear cartilage in control group was transplanted directly into the left side of the superficial fascia lacuna on the back of the rabbit,the piece of ear cartilage in experimental group combine with PRF was transplanted into the right side of superficial fascia lacuna on the back of the rabbit.General observation,weight comparison and statistical analysis and staining which includ hematoxylin-eosin staining,saffron O staining and alcian blue staining were performed after 2 weeks,4 weeks,8 weeks and 12 weeks.Results General observation and comparison of cartilage weight showed that:the weight of ear cartilage increased at 2-4 weeks,and decreased at 8-12 weeks.At each observation time,the weight of ear cartilage in the experimental group was greater than that in the control group.And the difference between the two groups was statistically significant(P<0.05).Staining observation showed that:at each observation time,the chondrocytes in the experimental group were significantly more than those in the control group under the same microscopic scale and the same visual field.The heterochromatic cartilage matrix and blue glycosaminoglycan were more obvious in the experimental group.Conclusion Autologous platelet-rich fibrin can promote the survival of rabbit ear cartilage after transplantation.
