摘要
目的探讨金丝桃苷(Hyperoside,HS)对脂多糖(Lipopolysaccharide,LPS)诱导体外SD大鼠软骨细胞骨关节炎模型的作用。方法选取5 d的SD大鼠膝关节软骨细胞,以LPS诱导的细胞炎症模型分为空白对照(Control)组,LPS诱导处理(Osteoarthritis,OA)组,LPS诱导和HS处理(OA+HS)组,分别处理24 h后,采用番红O染色观察细胞形态和细胞外基质分泌情况,以荧光染色检测观察细胞的活性,采用实时荧光定量PCR(RT-qPCR)技术检测软骨细胞炎症相关基因(TNF-a、MMP3、MMP13、IL-6)的表达量。结果HS能够维持软骨细胞的形态,提高细胞外基质的表达,降低细胞死亡率,维持细胞的活性;且HS能显著降低MMP3、TNF-a等多个炎症相关基因的表达水平(P<0.05)。结论HS能减少SD大鼠软骨细胞的细胞基质丢失,维持细胞活性,并能降低细胞炎症相关基因的表达,减少炎症的发生,对体外骨关节炎具有一定的治疗作用。
Objective To explore the effect of hyperoside(HS)on chondrocytes induced by lipopolysaccharide(LPS)in vitro.Methods Chondrocytes were extracted 5-days-old SD rats and divided into blank control group,lipopolysaccharide(OA)group,lipopolysaccharide plus hyperoside(OA+HS)group.After treatment for 24 h,Safranin O staining and fluorescent staining were used to detected the expression of the cell matrix and cell activity.The total RNA of chondrocytes was extracted,and then the expression of inflammation-related genes was detected by RT-qPCR.Results HS improved the expression of cell matrix compared to OA group,and higher cell viability was tested in the OA+HS group.HS also down-regulated the expression of inflammation-related genes(TNF-a、MMP3、MMP13、IL-6)(P<0.05 for each),which were significantly lower than those of the OA group.Conclusion HS can reduce the loss of cell matrix and maintain cellular activity of chondrocytes,and down-regulated the expression of cell inflammation-related genes,which may have a therapeutic effect on osteoarthritis.
