摘要
目的通过TCGA、GEO、Oncomine等公开数据库探讨ST3GAL5在膀胱尿路上皮癌患者中的表达水平、预后特征以及作用机制。方法收集TCGA、GEO、Oncomine数据库中关于ST3GAL5在膀胱尿路上皮癌中的表达谱,使用R语言分别研究ST3GAL5在膀胱尿路上皮癌与正常膀胱、肌层浸润性膀胱癌与非肌层浸润性膀胱癌以及高级别膀胱尿路上皮癌与低级别膀胱尿路上皮癌中的表达水平,绘制Kaplan-Meier曲线以评估ST3GAL5表达与总体生存率和复发的关系,并对ST3GAL5在膀胱尿路上皮癌中的信号通路作基因集富集分析。结果与正常膀胱组织比较,ST3GAL5在膀胱癌中表达下调,在肌层浸润性膀胱癌及高级别膀胱癌中ST3GAL5表达均显著下调。生存分析提示,在总体生存率及无复发生存率方面,ST3GAL5低表达比高表达均具更高的风险。结论ST3GAL5可能是膀胱癌的抑癌基因,ST3GAL5高表达可能抑制膀胱癌向肌层侵犯和高级别进展,因此,有可能成为肿瘤治疗的分子靶点。
Objective To explore the expression,prognosis and mechanism of ST3GAL5 in bladder urothelial carcinoma(BLCA)patients by TCGA,GEO,Oncomine and other public databases.Methods The expression profile of ST3GAL5 in BLCA were obtained from TCGA,GEO,and Oncomine databases.We investigated ST3GAL5 expression level between BLCA and normal bladder tissues,muscle invasive bladder cancer(MIBC)and non-muscle invasive bladder cancer(NMIBC),high grade BLCA and low grade BLCA,respectively using R packages.The Kaplan-Meier survival curves were generated to estimate the association between ST3GAL5 expression and overall survival status(OSS)and recurrence status(RS)in BLCA patients.Finally,gene set enrichment analysis(GSEA)was performed to identify the signaling pathways between high and low expression of ST3GAL5 in BLCA.Results Compared with normal bladder,ST3GAL5 expression was significantly lower in BLCA,and ST3GAL5 expression in MIBC and high grade BLCA were significantly lower than NMIBC and low grade BLCA,respectively.The survival analysis showed that lower expression of ST3GAL5 had a higher risk in terms of OSS and RS than higher expression of ST3GAL5 in BLCA.Conclusions ST3GAL5 may be an anti-oncogene in BLCA,so it may be a potential predictive and prognostic marker for MIBC and high grade BLCA,and even a molecular target for tumor therapy.
