摘要
目的探究白细胞介素(IL)-17对血管内皮细胞的作用,并观察紫草素对IL-17作用的干预。方法细胞实验:在人脐静脉内皮细胞株(HUVEC)中加入IL-17(100 ng/mL),观察IL-17对HUVEC成管的影响;动物实验:将15只小鼠随机分为5组:空白对照组,模型组,甲氨喋呤(MTX)组,紫草素低剂量及紫草素高剂量组。利用小鼠基质胶栓模型模拟皮下环境,并予各组灌胃1次/d持续12 d,空白对照及模型组予基质油[2.5%二甲基亚砜(DMSO),200μL/d],MTX组予MTX[0.5 mg/(kg·d)],紫草素低剂量组予Alkannin(2.5 mg/kg·d),紫草素高剂量组予Alkannin[5 mg/(kg·d)]。第14日处死小鼠,HE染色观察病理现象,免疫组化染色比较血管内皮细胞表面标志物CD34、血管内皮生长因子(VEGF)的表达。结果细胞实验:与对照组相比,IL-17组HUVEC成管能力显著提高;动物实验:模型组胶栓内血管内皮细胞数量较多,有微血管形成且VEGF表达升高,与之相比,紫草素各组胶栓内血管内皮细胞数量、微血管形成及VEGF表达量均下降且高剂量组更明显。结论紫草素可以显著抑制IL-17对血管内皮细胞迁移及成管的诱导作用。
Objective In order to investigate the effects of interleukin(IL)-17 on vascular endothelial cells and the effects of Alkannin on IL-17.Methods Cells:IL-17(100 ng/mL)was added into human venous endothelial cells(HUVEC)to observe its influence on HUVEC.Animals:15 mice were randomly divided into five groups including control group,model group,Methotrexate(MTX)group,Alkannin low-dose group and high-dose group.Mouse matrigel plug assay was used to imitate the subcutaneous tissue.Each group was given drugs once a day for 12 days.Control and model groups were given matrix oil(2.5%DMSO,200μL/d),and MTX group was given MTX(0.5 mg/kg·d).Alkannin groups were given Alkannin(2.5 mg/kg·d,5 mg/kg·d,respectively).All mice were sacrificed on the 14th day.Hematoxylin and eosin(HE)staining was used to observe pathological phenomenon,and immunohistochemical staining was used to test the expression of CD34 and vascular endothelial growth factor(VEGF).Results Compared with control group,tube formation of HUVEC significantly improved in IL-17 group.In animal models,there were more vascular endothelial cells in the matrigel plugs of model group in which microvessel formation and VEGF expression were increased compared with control group.Notably,the number of vascular endothelial cells and microangiogenesis as well as the expression of VEGF of Alkannin groups decreased,which were more profound in the high-dose group.Conclusion Alkannin can significantly inhibit IL-17-induced vascular endothelial cell migration and tube formation.
