摘要
目的利用F基因型腮腺炎病毒浓缩后疫苗原液,经不同途径免疫动物制备抗腮腺炎病毒血清。方法将F基因型腮腺炎病毒浓缩后疫苗原液与弗氏佐剂混合后,经皮下注射、肌肉注射及皮下+肌肉注射的三种途径免疫雌性家兔和雌性豚鼠,均于0、7、14、28和35 d免疫,每次7.5 lgCCID50/mL,于每次免疫后7 d采用微量滴定法检测血清抗体效价。利用抗体效价较高血清对麻疹病毒、风疹病毒和腮腺炎病毒进行干扰试验。结果不同免疫途径均可诱导动物产生抗体,皮下+肌肉注射的免疫方式产生的血清抗体效价高于单独皮下及肌肉注射的免疫方式,差异有统计学意义(P<0.05);在同等免疫剂量下,家兔产生的抗血清效价明显高于豚鼠。家兔抗腮腺炎病毒血清对麻疹病毒、风疹病毒和腮腺炎病毒的滴度均无干扰。结论经皮下和肌肉注射结合的途径免疫家兔,可获得高效价血清抗体,对制备高效价F基因型腮腺炎病毒血清抗体提供了依据。
Objective To prepare the antisera against mumps virus genotype F by immunization animals with concentrated bulk of vaccine by various routes.Methods Concentrated bulk of vaccine against mumps virus genotype F was mixed with Freund adjuvant,with which female rabbits and female guinea pigs were immunized by subcutaneous,intramuscular and subcutaneous+intramuscular routes respectively at a dosage of 7.5 lgCCID50/mL on days 0,7,14,28 and 35.Serum samples were collected 7 d after each dose and determined for antibody titer by microtitration method.Interference tests on measles,rubella and mumps viruses were carried out with the high titer antisera.Results Antisera were induced by various routes.The titer of antisera induced by subcutaneous+intramuscular route was significantly higher than those by subcutaneous or intramuscular route alone(P<0.05).However,the titer of antisera induced in rabbits was significantly higher than that in guinea pigs immunized at the same dosage.Rabbit antisera against mumps showed no interference to the titers of measles,rubella and mumps viruses.Conclusion High titer antiserum was prepared by immunization of rabbits by subcutaneous+intramuscular routes,which provided a basis for the preparation of rabbit antiserum against mumps virus of genotype F.
