摘要
建立头孢噻呋钠(ceftiofur sodium,CS)联合脂多糖(lipopolysaccharide,LPS)诱发鸡实验性肝损伤模型。选取1日龄健康海蓝褐蛋鸡140羽,依次开展头孢噻呋钠与LPS单用和联用致鸡肝损伤试验,以鸡血清中谷丙转氨酶(alanine transaminase,ALT)、谷草转氨酶(aspartate aminotransferase,AST)和肝脏病理学变化为肝损伤判断依据。结果显示,不同剂量头孢噻呋钠联合LPS(4 mL/kg)模型组ALT和AST显著升高(P<0.05),出现不同程度肝细胞病变;固定头孢噻呋钠剂量(5 mg/kg),当LPS剂量为2、4、6 mL/kg时,ALT和AST显著升高(P<0.05),LPS(2 mL/kg)组肝细胞损伤轻,LPS(4 mL/kg)组肝细胞发生明显空泡变性与脂肪变性,LPS(6 mL/kg)组肝细胞坏死严重,鸡只部分出现死亡。可见,单用LPS可造成肝损伤,但头孢噻呋钠与LPS联用引发肝损伤程度高于单用LPS。研究结果表明,其造模方法为给鸡皮下注射头孢噻呋钠(5 mg/kg),1次/d,连续3 d,第3天皮下注射完后再腹腔注射LPS(4 mL/kg),可诱导鸡实验性肝损伤。
To establish the experimental liver injury chicken model induced by ceftiofur sodium(CS)combined with lipopolysaccharide(LPS),140 healthy one-day-old Hai-lan-he chickens were selected.The dose screening test of CS inducing liver injury in chickens,LPS induced chicken liver injury dose screening test and a comparative study of LPS used only or combined with CS in the process of inducing liver injury in chickens were carried out successively.The changes of alanine transaminase(ALT),aspartate aminotransferase(AST)in chicken serum and liver pathology were used as the judgment basis of liver injury.The results showed ALT and AST in serum significantly increased(P<0.05)in chickens with different doses of CS and LPS(4 mL/kg),and there were different degrees of hepatocellular lesions.When the dose of LPS was 2 mL/kg,4 mL/kg,and CS 5 mg/kg,ALT and AST in chicken serum were significantly increased(P<0.05).When LPS was 2 mL/kg,liver cell damage was mild.When LPS was 4 mL/kg,there were obvious vacuolation and steatosis in hepatocytes.When LPS was 6 mL/kg,liver cell necrosis was severe,there were no complete cells basically,and some chickens were dead.In addition,The results showed LPS alone can cause a certain degree of liver injury,but the combined use of CS and LPS can cause a higher degree of liver injury than LPS alone.The results showed that intraperitoneal injection of LPS(4 mL/kg)after subcutaneous injection of ceftiofur(5 mg/kg),once a day for 3 days,could induce experimental liver injury in chickens.
