摘要
[目的]采用超高效液相色谱-四级杆-飞行时间质谱(UPLC/Q-TOF MS/MS)技术,对稳心颗粒中的化学成分进行初步分析。[方法]采用ACQUITY UPLC BEH C18色谱柱(2.1 mm×100 mm,1.7μm),以乙腈-水(含体积比0.1%甲酸)为流动相梯度洗脱,流速为0.3 mL/min,电喷雾电离源,正/负离子双模式检测,通过UPLC/Q-TOF MS/MS所得到的化合物分子量以及MS/MS碰撞解离碎片,结合文献报道,对稳心颗粒的主要化学成分进行分析鉴定。[结果]结合文献报道和一级MS、二级MS/MS高分辨质谱数据,对其中68个化合物进行了初步鉴定,结果显示,稳心颗粒成分以糖类、皂苷类、黄酮类为主,其中三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1丰度较高。[结论]UPLC/Q-TOF MS/MS技术是鉴定中药复方成分的有效手段,本研究首次利用该技术对稳心颗粒的主要成分进行定性分析,为阐明该复方药效物质基础、探讨其作用机制及其质量控制提供了有效的科学依据。
[Objective]Preliminarily analyze the chemical constituents of Wenxin Granules by ultra-high performance liquid chromatography-tandem quadrupole time of flight mass spectrometry(UPLC/Q-TOF-MS).[Methods]An ACQUITY UPLC BEH C18 column(2.1 mm×100 mm,1.7μm)was used,with acetonitrile-water(containing 0.1%formic acid)as mobile phase gradient elution,flow rate 0.3 mL/min,electrospray ionization source(ESI source),positive/negative ion(Positive/Negative)dual mode detection.The molecular weights of compounds obtained by UPLC/Q-TOF MS/MS and MS/MS collision dissociation fragments were used to analyze and identify the main chemical components of Wenxin Granules combined with literature report.[Results]Combined with literature reports and primary MS and secondary MS/MS high resolution mass spectrometry data,a preliminary identification of 68 of these compounds was performed.The results showed that the main components of Wenxin Granules were sugars,saponins,flavonoids,among which,notoginsenoside R1,ginsenoside Rg1,Rb1 are abundant.[Conclusion]UPLC/Q-TOF MS/MS is an effective method to identify the components of traditional Chinese medicine compound.This study is the first time to use this technology to carry out qualitative analysis on the main components of Wenxin Granules,which provides an effective scientific basis for clarifying the material basis of the efficacy of compound Chinese medicine,exploring its mechanism of action and quality control.
