摘要
以甲基营养型芽孢杆菌(Bacillus methylotrophicus)BMF 04为试材,采用平板透明圈法、比色法和盆栽试验,研究海洋细菌BMF 04发酵液不同处理方法对黄瓜幼苗生长的影响,测定菌株的固氮、解磷、解钾作用以及产IAA、铁载体、ACC脱氨酶能力,以期明确BMF04菌株的促生作用及其机理。结果表明:BMF 04菌株发酵液处理的黄瓜种子及其栽培土壤,均能显著提高黄瓜的出苗率、地上部和地下部鲜质量、茎粗、株高和须根数,其中拌土处理效果最好,三叶期幼苗茎粗为4.7 mm,株高为12.4 cm,单株地上部鲜质量和地下部鲜质量分别为9.49、0.95 g,单株须根数152根;BMF 04菌株具有固氮作用,可以产生ACC脱氨酶、IAA和铁载体,其中铁载体和IAA产生量随着培养时间的延长逐渐增加,24 h产铁载体的相对含量达到最高为34.82%,32 h IAA的分泌量达到最大值为2.647 mg·L-1,随后二者的分泌量逐渐减少;该菌株不具有解磷和解钾能力。
Taking Bacillus methylotrophicus BMF 04 as the test material,using plate transparent circle method,colorimetry and pot experiment,to clarify the growth promoting effect and mechanism of BMF 04 strain,the effects of different treatment methods of marine bacteria BMF 04 fermentation broth on cucumber seedling growth,nitrogen fixation,ability in decomposing of phosphate and silicate,production of IAA,siderophore and ACC deaminase activity of the strain were studied.The results showed that the cucumber seeds treated with BMF 04 strain fermentation broth and its cultivation soil could significantly improve the emergence rate,fresh weight of single plant abaveground and underground,stem diameter and length,number of roots single,of which the best soil mixing method,three-leaf seedling stem diameter 4.7 mm,plant height of 12.4 cm,the fresh weight of the aboveground part and the fresh weight of the underground part of a single plant were 9.49 g and 0.95 g respectively,number of roots was 152 radical per plant;BMF 04 strain had the effect of nitrogen fixation,could produce IAA,siderophore and ACC deaminase,where in the siderophore and IAA production with the extension of the incubation time gradually increased,the relative content of siderophore reached 34.82% at 24 hours,and the secretion of 32 hours IAA reached 2.647 mg·L-1 at the maximum,then the secretion of both gradually decreased;the strain did not had the ability in decomposing of phosphate and silicate.
