摘要
目的基于网络药理学方法探讨加味茵陈四逆汤(JWYCSNT)干预肝纤维化/肝硬化的作用靶点及通路,预测其潜在作用机制。方法基于TCMSP数据库提取加味茵陈四逆汤的有效活性成分及作用靶点,通过人类基因数据库(GeneCards)获取肝纤维化/肝硬化治疗靶点,并利用R语言筛选共同靶点;使用Cytoscape软件构建"药物有效活性成分-疾病靶点"可视化调控网络图,利用STRING数据库构建蛋白互作网络图(PPI),然后运行R语言对PPI进行核心蛋白的筛选;最后,通过R语言对共同靶点进行基因本体(GO)功能富集分析和京都基因与基因组百科全书(KEGG)通路富集分析。结果共获得132个有效活性成分,93个共同靶点;核心蛋白主要包括MAPK8/JNK、PPARG、RELA/NF-κB等;GO功能富集分析涉及101个,影响核受体活性、基因转录等过程;KEGG通路富集分析得出114条通路,涉及细胞凋亡、乙型肝炎、丙型肝炎、肝细胞性肝癌等通路,涵盖了MAPK、NF-κB、PPAR、Wnt等肝纤维化重要通路。结论通过网络药理学的方法,证实加味茵陈四逆汤具有多成分、多靶点、多通路的作用特点,并预测其干预肝纤维化/肝硬化的潜在作用机制,可为基础及临床研究提供参考依据。
Objective To explore the targets and pathways of Jiaway Yinchen Sini(JWYCSNT) Decoction in the intervention of liver fibrosis/cirrhosis based on network pharmacology, and to predict its potential mechanism.Methods Based on the TCMSP database,the effective active components and targets of the Jiaway Yinchen Sini Decoction were extracted,and the targes of liver fibrosis/cirrhosis were obtained through the human gene database(GeneCards);and then the common targets were screened by R language. Cytoscape software was engaged to construct a visual regulatory network map of "drug-active ingredient-disease-target", and a protein-protein interaction network map(PPI)was constructed using the STRING database. Then the R language was used to screen the core protein of the PPI;finally,the R language performed functional gene ontology(GO)enrichment analysis and common enrichment analysis of the Kyoto Gene and Genomic Encyclopedia(KEGG) pathways for common targets. Results A total of 132 active components and 93 common targets were obtained. The core proteins mainly included MAPK8/JNK, PPARG, and RELA/NF-κB, etc. The GO functional enrichment analysis involved 101 genes,involving nuclear receptor activity and gene transcription,etc. KEGG pathway enrichment analysis resulted in 114 pathways involving apoptosis,hepatitis B,hepatitis C,hepatocellular carcinoma,etc.,covering MAPK,NF-κB, PPAR, Wnt and other important liver fibrosis related pathways. Conclusion This study confirmed the multi-component, multi-target and multi-channel characteristics of Jiawei Yinchen Sini Decoction by network pharmacology, and predicted its potential mechanism of intervention in liver fibrosis/cirrhosis, also provide a reference for basic and clinical research.
