摘要
目的:建立高效液相色谱法同时测定妇炎舒胶囊中红景天苷、绿原酸、芍药苷、虎杖苷、白藜芦醇、延胡索乙素和丹酚酸B的含量。方法:应用HPLC多波长切换梯度洗脱技术,采用Kromasil C18(4.6 mm×250 mm,5μm)色谱柱,以乙腈(A)-0.1%磷酸溶液(B)为流动相,流速:1.0 ml·min^-1,检测波长:280 nm(红景天苷、延胡索乙素、丹酚酸B)、327 nm(绿原酸)、230nm(芍药苷)、306 nm(虎杖苷、白藜芦醇),柱温:30℃,进样量:10μl。结果:红景天苷、绿原酸、芍药苷、虎杖苷、白藜芦醇、延胡索乙素和丹酚酸B检测质量浓度的线性范围分别为3.842~69.156,2.536~40.167,5.073~91.314,1.602~28.836,0.553~9.954,1.923~34.605,1.344~24.192μg·ml^-1(r为0.9993~0.9999);平均加样回收率分别为98.9%、100.3%、99.8%、99.6%、98.3%、100.5%、100.8%(RSD<2.0%,n=6);精密度、重复性、稳定性(24 h)试验的RSD<2.0%(n=6)。结论:本研究建立的方法可用于妇炎舒胶囊的多组分同时测定。
Objective:To develop a method for the simultaneous determination of salidroside,chlorogenic acid,paeoniflorin,polydatin,resveratrol,tetrahydropalmatine and salvianolic acid B in Fuyanshu capsules by HPLC.Methods:The determination was carried out on a Kromasil C18(4.6 mm×250 mm,5μm)column with the mobile phase consisting of acetonitrile(A)-0.1%phosphoric acid(B)in a gradient mode at a flow rate of 1.0 ml·min^-1.The detection wavelength was set at 280 nm for salidroside,tetrahydropalmatine and salvianolic acid B,327 nm for chlorogenic acid,230 nm for paeoniflorin,and 306 nm for polydatin and resveratrol.The column temperature was set at 30℃and the injection volume was 10μl.Results:The linear range was 3.842-69.156μg·ml^-1 for salidroside,2.536-40.167μg·ml^-1 for chlorogenic acid,5.073-91.314μg·ml^-1 for paeoniflorin,1.602-28.836μg·ml^-1 for polydatin,0.553-9.954μg·ml^-1 for resveratrol,1.923-34.605μg·ml^-1 for tetrahydropalmatine,and 1.344-24.192μg·ml^-1 for salvianolic acid B(r=0.9993-0.9999);the average recovery was 98.9%,100.3%,99.8%,99.6%,98.3%,100.5%and 100.8%(RSD<2.0%,n=6),respectively.The RSDs of precision,reproducibility and stability tests(24 h)were lower than 2.0%(n=6).Conclusion:The method can be used to determine multi-constituents simultaneously in Fuyanshu capsules.
