附子总碱对大鼠心功能及离体心脏的损害作用及其机制 被引量:5

Effects of Aconite Total Alkaloids on Cardiac Function and Isolated Heart in Rats and Its Mechanism

摘要 为了探讨附子总碱对大鼠心功能、离体心脏的毒性作用及其作用机制,本研究选取50只Wistar大鼠,采用随机数字表法分为A组(等量生理盐水灌胃)、B组(附子药液提取物2.72 g/kg灌胃)、C组(附子药液提取物3.68 g/kg灌胃)、D组(附子药液提取物4.28 g/kg灌胃)、E组(附子药液提取物4.98 g/kg灌胃)各10只,每天灌胃1次,连续3 d后分别取静脉血清备用:(1)取出大鼠心脏后分别给予不同组别的含药血清实施Langedorff离体心脏灌流实验,记录给药5 min后各组离体心脏的左心室收缩压(LVSP)、左心室舒张末期压(LVEDP)、左心室内压最大上升速率(+dp/dt)、左心室内压最大下降速率(+dp/dt)、心输出量(CO)、心率(HR);(2)采用不同组别的含药血清Wistar大鼠心肌细胞,检测各组上清液中丙二醛(MDA)含量、心肌细胞的存活率。A组、B组、C组、D组大鼠离体心脏的HR差异无统计学意义(p>0.05),E组的大鼠离体心脏的HR显著低于A组、B组、C组、D组(p<0.05);A组、B组、C组、D组大鼠离体心脏的CO值均逐渐升高(p<0.05),E组大鼠离体心脏CO值较D组减小(p<0.05);A组、B组、C组、D组大鼠离体心脏的LVSP、+dp/dt、-dp/dt呈逐渐升高的趋势,组间比较差异均具有统计学意义(p<0.05),E组的大鼠离体心脏的LVSP、+dp/dt、-dp/dt低于A组、B组、C组、D组(p<0.05);A组、B组、C组、D组大鼠离体心脏的LVEDP差异不具有统计学意义(p>0.05),E组大鼠离体心脏的LVEDP较D组减小(p<0.05);A组、B组、C组、D组含药血清培养的心肌细胞上清液中MDA测定值比较,差异均不具有统计学意义(p>0.05),E组含药血清培养的心肌细胞上清液中MDA测定值高于A组、B组、C组、D组(p<0.05);A组、B组、C组、D组含药血清培养的心肌细胞活性差异不具有统计学意义(p>0.05),E组含药血清培养的心肌细胞活性低于A组、B组、C组、D组(p<0.05)。低剂量的附子总碱对大鼠心功能、离体心脏均具有一定的正性促进作用,但是随着剂量的增加,其对心脏的毒性作用将逐渐明显。 To investigate the toxic effects of aconite total alkaloids on cardiac function and isolated hearts in rats and its mechanism.50 Wistar rats were randomly divided into group A(equal saline),group B(2.72 g/kg of aconite solution),and group C(aconite extract 3.68 g/kg gavage),group D(Aconite liquid extract 4.28 g/kg gavage),group E(Aconite solution extract 4.98 g/kg gavage)each 10,once a day,The serum was taken for 1 times per day for 3 days,and the venous serum was taken for later use:(1)After taking out the rat heart,Langedorff isolated heart perfusion experiments were performed with different groups of drug-containing serum,and the left ventricular systolic pressure(LVSP)of each group was recorded after 5 minutes of administration.Left ventricular end-diastolic pressure(LVEDP),maximum rate of increase of left ventricular pressure(+dp/dt),maximum rate of left ventricular pressure(+dp/dt),cardiac output(CO),heart rate(HR);(2)Different groups of drug-containing serum Wistar rat cardiomyocytes were used for 24 hours to detect the content of malondialdehyde(MDA)and the survival rate of cardiomyocytes in the supernatant of each group.There was no significant difference in HR between the rats in group A,group B,group C and group D(p>0.05).The HR of isolated rats in group E was significantly lower than that in group A,group B group C and group D(p<0.05);CO values of isolated hearts of rats in group A,group B,group C and group D increased gradually(p<0.05).The CO value of isolated heart in group E was higher than that in group D.The decrease(p<0.05);The LVSP,+dp/dt,and-dp/dt of the isolated hearts of group A,B,C and D showed a trend of increasing,and the differences between the groups were statistically significant.The significance of learning(p<0.05),LVSP,+dp/dt,-dp/dt of isolated rat hearts in group E were lower than those in group A,group B,group C and group D(p<0.05);Group A,There was no significant difference in LVEDP between the B group,C group and D group(p>0.05).The LVEDP of the isolated group was significantly lower than that of the D group(p<0.05).Group A,The MDA values in the supernatants of the cultured serum of group B,C and D were not statistically significant(p>0.05),and the supernatant of cardiomyocytes cultured in serum containing group E was in the supernatant.The measured value of MDA was higher than that of group A,group B,group C and group D(p<0.05);Group A,group B,group C and group D were cultured with drug-containing serum.Myocyte activity differences were not statistically significant(p>0.05),serum drug containing cultured myocardial cells group E activity was lower than in group A,group B,group C and D(p<0.05).The low dose of aconite total alkaloid has a positive effect on the heart function and isolated heart of rats,but with the increase of dose,its toxic effect on the heart will gradually become obvious.
