人外周血染色体核型制备改良方法探讨 被引量:8

摘要 目的探讨改良人外周血染色体细胞培养和制备条件对外周血染色体核型的影响。方法在传统方法基础上,通过细胞培养阶段延长外周血细胞培养时间,细胞收获阶段增加秋水仙碱浓度和减少秋水仙碱作用时间来改良外周血染色体核型制备技术,对改良前和改良后不同方法对各200例标本的外周血染色体核型结果进行统计分析,研究改良方法对外周血染色体核型结果的影响。结果统计结果显示改良前200份样本获得的分裂相总数(25371个)明显少于改良后获得的分裂相总数(61810个),且可用于分析的优质核型数(1038个)要显著低于改良后的优质核型数(2210个),上述结果具有统计学差异意义。结论通过在细胞培养阶段增加外周血细胞培养时间,在细胞收获阶段增加秋水仙碱浓度,并同时缩短秋水仙碱的作用时间可有效获得更多显带清晰、结果稳定的外周血染色体核型,并改进外周血细胞染色体核型质量,且改进方法可以节约细胞收获和染色体标本制备的时间。改良方法具有质量稳定、操作简便、可推广应用等特点。 Objective To explore the effects of modified factors on affecting the karyotype of human peripheral blood.Methods Several factors were changed during the process for preparing the karyotype of human peripheral blood,such as prolonging the culture time for the peripheral blood cells,increasing the concentration of colchicines and reducing the time of colchicines on affecting the cells.Results Totally,25371 karyotypes were captured for 200 samples cultured 72 hours,which was less than that of 61810 karyotypes for 200 samples by prolonging the culture time to 96 hours(Z=-13.421,P=0.000,P<0.05).And 1038 metaphase chromosome and 2210 metaphase chromosome with higher resolution G band were observed from 200 samples,respectively,we got higher number of metaphase chromosome with clear G band with the improved method than traditional method.Further statistical analysis revealed the significantly differences between two groups(Z=-12.493,P=0.000,P<0.05).Conclusion The improved method can effectively obtain the higher resolution chromosomal karyotypes of peripheral blood and effectively reduce the time of bench work.
