摘要
采用离子液体([BMIM]pH6)辅助乙醇法提取山楂红色素,以离子液体用量、液料比、提取时间、溶液pH及提取温度为自变量,以吸光度为响应值,运用Plackett-Burman试验设计对自变量进行筛选;采用响应面分析法对山楂红色素提取工艺进行优化;采用液相色谱-质谱联用技术(UPLC-MS/MS)测定山楂红色素的成分组成。山楂红色素提取的最优条件:离子液体用量为15%,液料比为4.3mL/g,提取时间为59min,溶液pH为1.1,提取温度为60℃,并由此条件得到的吸光度为1.279。UPLC-MS测得花青素类花色苷和花翠素类花色苷,分别占总花色苷量的76.17%和18.98%。
Ionic liquid ([BMIM] pH6) was used as a kind of assistant in the ethanol extraction of hawthorn red pigment. The ionic liquid dosage, liquid ratio, extraction time, solution pH and extraction temperature were selected as independent variables, and the absorbance was selected as the response value. The independent variables were selected by Plackett-Burman test. The extraction process was optimized by the response surface analysis. The composition of hawthorn red pigment was determined by Ultra Performance Liquid Chromatography-mass spectrometry (UPLC-MS/MS). The optimum extraction parameters were determined as follows : ionic liquid dosage, 15%, liquid ratio, 4.3 mL/g, extraction time, 59 min, pH, 1.1, extraction temperature, 60℃. And the absorbance obtained by these conditions was 1.279. Anthocyanins and delphinidin anthocyanins were determined by UPLC-MS/MS, which accounted for 76.17% and 18.98% of total anthocyanin, respectively.
