瑞舒伐他汀对心肌缺血再灌注损伤大鼠心肌细胞及氧化应激反应的影响 被引量:2

Effects of rosuvastatin on myocardial cells and oxidative stress in rats with myocardial ischemia-reperfusion injury

摘要 目的研究瑞舒伐他汀对心肌缺血再灌注损伤(MIRI)大鼠心肌细胞及氧化应激反应的影响。方法选择健康Wistar大鼠60只,随机分为假手术组、MIRI组、MIRI+瑞舒伐他汀低、中、高剂量组,每组12只,雌雄各6只。MIRI组、MIRI+瑞舒伐他汀低、中、高剂量组采用结扎冠状动脉左前降支的方法建立MIRI模型,建模前连续7d给予MIRI+瑞舒伐他汀低、中、高剂量组大鼠瑞舒伐他汀0.5、1.0、2.0mg/kg灌胃,假手术组、MIRI组给予同等体积生理盐水灌胃。比较各组大鼠血清心肌酶水平及心肌组织中氧化应激指标水平。结果MIRI组、MIRI+瑞舒伐他汀低、中、高剂量组血清AST、LDH、CK、CK-MB、α-HBD及MDA水平均高于假手术组,心肌组织SOD、GSH-Px、CAT、T-AOC及NOS水平均低于假手术组,且MIRI+瑞舒伐他汀低、中、高剂量组上述指标水平均明显优于MIRI组(P<0.05),且MIRI+瑞舒伐他汀低、中、高剂量组血清AST、LDH、CK、CK-MB、α-HBD及MDA水平逐渐降低,心肌组织SOD、GSH-Px、CAT、T-AOC及NOS水平逐渐升高。结论瑞舒伐他汀可有效减轻MIRI大鼠的氧化应激反应,减少缺血再灌注时心肌细胞的损伤。 Objective To study the effects of rosuvastatin on myocardial cells and oxidative stress in rats with myocardial ischemia-reperfusion injury (MIRI). Methods Sixty healthy Wistar rats were selected and randomly divided into sham-operated group, MIRI group, MIRI+rosuvastatin low, medium and high dose groups, with 12 rats in each group, 6 rats in male and 6 rats in female. MIRI model was established by ligation of left anterior descending coronary artery in MIRI group, MIRI + rosuvastatin low, medium and high dose groups. Before modeling, rats in the MIRI+rosuvastatin low, medium and high dose groups were given 0.5, 1.0 and 2.0 mg/kg of rosuvastatin intragastric administration for 7 conseautive days, and rats in the sham-operated group and the MIRI group were given the same volume of normal saline intragastric administration. The levels of serum myocardial enzymes and oxidative stress indexes in myocardium of rats in each group were compared. Results The levels of serum AST, LDH, CK, CK-MB,α-HBD and MDA in the MIRI group, MIRI+rosuvastatin low, medium and high dose groups were higher than those in the sham-operation group, and the levels of SOD, GSH-Px, CAT, T-AOC and NOS in myocardium were lower than those in the sham-operation group, the levels of above indicators in the MIRI+rosuvastatin low, medium and high dose groups were better than those in the MIRI group (P<0.05), and the levels of serum AST, LDH, CK, CK-MB,α-HBD and MDA in the MIRI+rosuvastatin low, medium and high dose groups decreased gradually, the levels of SOD, GSH-Px, CAT, T-AOC and NOS in myocardium increased gradually. Conclusion Rosuvastatin can effectively alleviate oxidative stress in MIRI rats, and reduce myocardial cell injury during ischemia-reperfusion.
