摘要
研究超声波辅助乙醇提取薤白皂苷的最优工艺条件,并对其抑菌活性进行研究。以干燥后的薤白为原料,在单因素实验的基础上,选取固液比、超声功率、提取时间为自变量,皂苷提取率为响应值,根据响应面BoxBehnken试验设计原理,采用三因素三水平的分析方法模拟得到二次多项回归方程。同时采用抑菌圈法测定不同浓度薤白皂苷的抑菌活性。研究发现回归模型具有显著性,方程拟合较好,可对薤白皂苷提取率进行预测:即固液比1∶50,提取温度64℃,超声功率500 W,在此条件下薤白皂苷的提取率可达4.86%。研究并发现薤白皂苷对供试微生物都有一定的抑制效果,抑菌效果大小顺序为大肠杆菌、金黄色葡萄球菌、枯草芽孢杆菌、酿酒酵母、黑曲霉和桔青霉。
The objective of this study was to optimize the ultrasound-assisted ethanol extraction of saponin from A. macrostemon and evaluale its antimicrobial activity.The dry powder of bulbs was used as the material. On the basis of single-factor test,the response surface methodology was used to investigate the effects of pro- cessing parameters,including solvent/material ration,ultrasound power and temperature.The extraction rate of saponin was chosen as the response value,the mathematical model was established by implementing three factors and three levels,the antibacterial effects of the extract from A. macrostemon were investigated.The results showd that the optimal process parameters were as follows:solvent/material ratio 1∶50,extracted temperature 64 ℃,ultrasound powder 500 W,the extraction ration of A. macrostemon was up to 4.86%,and experimental values coincided well with the predicted values.The results indicated that the order of the antimicrobial susceptibility of the extract was:E. coli>S. aureus>B.subtilis>A. cerevisia>A. niger>Penicillium sp..
